【摘要】：目的:研究鼻敏感顆粒對變應性鼻炎小鼠樹突狀細胞(DC)發(fā)揮免疫調節(jié)作用的具體機制。方法:分離出小鼠骨髓細胞,使用rGM-CSF和rIL-4共培養(yǎng)骨髓細胞,刺激骨髓細胞向DC轉化。然后加入脂多糖和不同濃度的鼻敏感顆粒共刺激未成熟DC,12h后檢測DC中MHCⅡ、CD80、CD86表面分子以及細胞上清中TNF-α、IL-1β、IL-6和IL-10的表達。結果:鼻敏感顆粒能夠顯著抑制成熟DC標志性細胞因子IL-6、TNF-α、IL-10、IL-1β的生成和成熟DC標志物MHCⅡ、CD80和CD86的表達。結論:鼻敏感顆粒能夠顯著抑制未成熟DC向成熟DC的分化。
