綠茶多酚對(duì)人肝癌細(xì)胞低密度脂蛋白受體功能的影響及機(jī)制

【摘要】:【目的】探討綠茶多酚主要活性成分表沒食子兒茶素沒食子酸酯(EGCG)對(duì)人肝癌細(xì)胞HepG2低密度脂蛋白受體(LDLR)功能的影響及其機(jī)制!痉椒ā坑肅CK8試劑盒檢測EGCG對(duì)HepG2細(xì)胞最適濃度及增殖活性的影響;用Western Blotting技術(shù)檢測HepG2細(xì)胞LDLR蛋白及前蛋白轉(zhuǎn)化酶枯草桿菌蛋白酶9(PCSK9)蛋白表達(dá)水平;用熒光顯微鏡觀察HpeG2細(xì)胞對(duì)低密度脂蛋白膽固醇(LDL)的攝取水平。【結(jié)果】EGCG可通過上調(diào)LDLR蛋白表達(dá)(P0.05),及下調(diào)PCSK9蛋白表達(dá)(P0.05),促進(jìn)HepG2細(xì)胞攝取LDL(P0.05)。【結(jié)論】EGCG可上調(diào)HepG2細(xì)胞LDLR蛋白表達(dá),其可能機(jī)制是通過下調(diào)HepG2細(xì)胞PCSK9蛋白的表達(dá),從而減少其對(duì)LDLR蛋白的降解,為冠心病降脂治療提供新的途徑。
[Abstract]:[objective] to investigate the effect of epigallocatechin gallate (EGCG), a major active component of green tea polyphenols, on the (LDLR) function of low density lipoprotein receptor (LDL) receptor in human hepatocellular carcinoma cell line HepG2 and its mechanism. [methods] CCK8 kit was used to detect the effect of epigallocatechin gallate. The effect of EGCG on the optimal concentration and proliferative activity of HepG2 cells; The expression levels of LDLR protein and PCSK9 protein in HepG2 cells were detected by Western Blotting technique. The uptake of low density lipoprotein cholesterol (LDL) in HpeG2 cells was observed by fluorescence microscope. [results] EGCG could up-regulate the expression of LDLR protein (P0.05) and down-regulate the expression of PCSK9 protein (P0.05). [conclusion] EGCG can up-regulate the expression of LDLR protein in HepG2 cells by down-regulating the expression of PCSK9 protein in HepG2 cells, thereby reducing the degradation of LDLR protein. [conclusion] EGCG can up-regulate the expression of LDLR protein in HepG2 cells (P0.05). It provides a new way for the treatment of coronary heart disease.
