雙氫楊梅樹皮素對(duì)人視網(wǎng)膜母細(xì)胞瘤細(xì)胞色素C氧化酶表達(dá)的影響

【摘要】:目的:測(cè)定有/無雙氫楊梅樹皮素(ampelopsin,APS)干預(yù)時(shí)視網(wǎng)膜母細(xì)胞瘤(retinoblastoma,RB)細(xì)胞中細(xì)胞色素C氧化酶(cytochrome coxidase,COX)蛋白的表達(dá)變化,,探討藤茶雙氫楊梅樹皮素對(duì)體外培養(yǎng)的人視網(wǎng)膜母細(xì)胞瘤細(xì)胞生長的影響及其抗腫瘤作用機(jī)制。 方法:取處于對(duì)數(shù)生長期的人RB細(xì)胞株(HXO-RB44)進(jìn)行體外培養(yǎng),設(shè)置APS藥物濃度三組:33.33(高)、14.84(中)、4.40(低)ug/mL,并設(shè)空白對(duì)照組及順鉑(2.00ug/mL)陽性對(duì)照組。在APS藥物干預(yù)體外培養(yǎng)RB細(xì)胞24h后,采用WESTBOLT法測(cè)定視網(wǎng)膜母細(xì)胞瘤細(xì)胞中細(xì)胞色素C氧化酶蛋白IV亞基表達(dá)的灰度值,并求出相對(duì)灰度值。 結(jié)果:不同藥物濃度組APS干預(yù)體外培養(yǎng)的RB細(xì)胞24h后,均能明顯降低細(xì)胞色素C氧化酶IV亞基的活性表達(dá),各APS藥物組與空白對(duì)照組、陽性對(duì)照組比較,差異均有統(tǒng)計(jì)學(xué)意義,其中4.40ug/mL藥物濃度組降低COX IV活性表達(dá)最為明顯,相對(duì)灰度值為0.112±0.032。各藥物組間并不呈現(xiàn)隨藥物濃度增加,COX IV活性表達(dá)降低越明顯的特征。 結(jié)論:1.APS能降低COX IV活性的表達(dá),導(dǎo)致COX不能正常裝配,功能下降,線粒體呼吸鏈電子傳遞受到抑制,進(jìn)而引起線粒體損傷,激活腫瘤細(xì)胞的凋亡進(jìn)程,達(dá)到抗腫瘤目的。2.COX IV活性的表達(dá)有其獨(dú)特的調(diào)控機(jī)制,且可能存在啟動(dòng)閾值,其活性表達(dá)并不隨APS藥物濃度升高而降低。
[Abstract]:Objective: to determine the expression of cytochrome C oxidase (cytochrome coxidase,COX) protein in retinoblastoma (retinoblastoma,RB) cells treated with or without dihydromyricetin (ampelopsin,APS). To investigate the effect of Dihydromyricetin on the growth of human retinoblastoma cells in vitro and its antitumor mechanism. Methods: the human RB cell line (HXO-RB44) at logarithmic growth stage was cultured in vitro. The concentration of APS was set up in three groups: 33.33 (high), 14.84 (middle), 4.40 (low) ug/mL, blank control group and 2.00ug/mL positive control group. The expression of cytochrome C oxidase protein IV subunit in retinoblastoma cells was measured by WESTBOLT method after 24 hours of intervention by APS drugs, and the relative gray scale was obtained. Results: the activity of cytochrome C oxidase IV subunit was significantly decreased in RB cells cultured in vitro after 24 hours of intervention by APS in different concentration groups. The difference was statistically significant between each APS drug group and the blank control group and the positive control group. The activity of COX IV was significantly decreased in 4.40ug/mL group, with a relative gray value of 0.112 鹵0.032. The expression of, COX IV activity decreased with the increase of drug concentration. Conclusion: 1.APS can reduce the expression of COX IV activity, leading to the COX not being assembled normally, the function decreasing, the electron transfer of mitochondrial respiratory chain being inhibited, and then causing mitochondrial damage and activating the apoptosis process of tumor cells. The expression of 2.COX IV activity has its own regulatory mechanism, and there may be a priming threshold. The expression of 2.COX IV activity does not decrease with the increase of APS concentration.
