大鼠回腸貯袋炎的腸屏障變化研究

【摘要】:目的:建立大鼠回腸貯袋炎模型,研究大鼠回腸貯袋炎的腸屏障變化,并探討其發(fā)生改變的機制。方法:利用顯微外科技術建立大鼠回腸貯袋模型,將模型大鼠隨機分為IPAA組(接受回腸貯袋手術,術后正常飲水)和DSS組(接受回腸貯袋手術,術后第30天始,自由飲用5%DSS溶液4天),并設假手術組(接受開關腹手術,術后正常飲水)作為對照。實驗期間觀察記錄各組大鼠體重變化、便血情況、糞便評分;術后第35天,處死全部大鼠,假手術組取末端回腸組織,IPAA組及DSS組取貯袋組織及貯袋近端回腸組織,HE染色進行組織學評分;免疫組化染色檢測組織MPO、occludin蛋白表達水平;留取血標本,采用比色法測定血D-乳酸濃度;實時熒光定量PCR檢測組織IL-1β、IL-6、TNF-α和IL-10的m RNA轉錄水平。結果:各組大鼠術后體重在短期下降之后逐漸增長,術后第30-35天,DSS組體重再次持續(xù)下降,假手術組和IPAA組則繼續(xù)增長;DSS組大鼠均出現(xiàn)血便(6/6),假手術組(0/6)和IPAA組(0/6)均無便血;DSS組糞便評分低于假手術組(P0.01)和IPAA組(P0.01),IPAA組糞便評分低于假手術組(P0.01);DSS組和IPAA組貯袋的組織學評分均高于DSS組回腸、IPAA組回腸和假手術組回腸(P0.01),且DSS組貯袋高于IPAA組貯袋(P0.01),而各組回腸的組織學評分之間差異無統(tǒng)計學意義(P0.05)。DSS組和IPAA組的MPO蛋白表達水平,血D-乳酸濃度,IL-1β、IL-6和TNF-α的m RNA轉錄水平均高于假手術組(P0.01),且DSS組均高于IPAA組(P0.01)。DSS組和IPAA組的occludin蛋白表達水平和IL-10m RNA轉錄水平均低于假手術組(P0.01),且DSS組均低于IPAA組(P0.01)。結論:1.成功建立大鼠回腸貯袋模型,并使用DSS成功誘導大鼠回腸貯袋炎模型。2.大鼠回腸貯袋炎模型的腸屏障結構和功能受損,受損程度和炎癥程度呈正相關。3.促炎因子相對增多和抗炎因子相對減少促進腸屏障受損。4.針對促炎因子、抗炎因子進行的治療,可能對腸屏障結構和功能的恢復有效。
[Abstract]:Aim: to establish a rat model of ileal pouch inflammation, to study the changes of intestinal barrier in rats with ileal pouch inflammation, and to explore the mechanism of the changes. Methods: rat ileal bag model was established by microsurgical technique. The model rats were randomly divided into two groups: IPAA group (receiving ileal bag operation, normal drinking water after operation) and DSS group (receiving ileal bag operation 30 days after operation). Free drinking of DSS solution for 4 days, and sham operation group (operation on switch abdomen, normal drinking water after operation) as control group. During the experiment, the changes of body weight, hematochezia and stool score were observed and recorded, and all the rats were killed on the 35th day after operation. In the sham operation group, the terminal ileum tissue of IPAA group and the DSS group were taken from the bag tissue and the proximal ileum tissue for histological grading; the expression of MPOOOOcludin protein was detected by immunohistochemical staining; the blood samples were collected. The serum Dlactic acid concentration was measured by colorimetry, and the mRNA transcription levels of IL-6TNF- 偽 and IL-10 were detected by real-time fluorescence quantitative PCR. Results: the weight of the rats in each group increased gradually after the short-term decrease, and the weight of the DSS group decreased again on the 30th to 35th day after operation. In the sham operation group and the IPAA group, the stool score in the DSS group was lower than that in the sham operation group and the IPAA group. The stool score in the DSS group was lower than that in the sham operation group (P 0.01) and the stool score in the IPAA group was lower than that in the sham operation group (P 0.01) and in the IPAA group (P 0.01 / 6). The stool score in the DSS group was lower than that in the sham operation group (P 0.01) and the stool score in the IPAA group was lower than that in the IPAA group. The histological scores of DSS group were higher than that of DSS group IPAA group and sham operation group (P 0.01), and that of DSS group was higher than that of IPAA group (P 0.01), but there was no significant difference in histological score between DSS group and IPAA group (P 0.05) and IPAA group (P < 0.05), the expression level of MPO protein in DSS group was higher than that in IPAA group, and the expression of MPO protein in DSS group was higher than that in IPAA group. The mRNA transcription levels of IL-1 尾 and TNF- 偽 in serum DLA were higher than those in sham operation group (P 0.01), and the levels of occludin protein expression and IL-10 mRNA transcription in DSS group were higher than those in IPAA group (P 0.01) and IPAA group (P 0.01), and the levels of IL-10 mRNA transcription in DSS group were all lower than those in IPAA group (P 0.01). Conclusion 1. The rat ileal bag model was successfully established and the rat ileal pouch model was successfully induced by DSS. The intestinal barrier structure and function of rat ileal pouch model were damaged, and the degree of injury was positively correlated with the degree of inflammation. The relative increase of proinflammatory factor and the decrease of anti-inflammatory factor promote intestinal barrier damage. 4. The treatment of proinflammatory factor and anti-inflammatory factor may be effective for the recovery of intestinal barrier structure and function.
