調節(jié)軟骨細胞骨架蛋白對成軟骨表型的影響

【摘要】:目的研究軟骨細胞骨架蛋白結構改變與成軟骨細胞表型改變的關系。方法采用三維微團培養(yǎng)法培養(yǎng)新西蘭兔膝關節(jié)軟骨細胞,將軟骨細胞分為對照組、秋水仙堿組、細胞松弛素B組及丙烯酰胺組,通過秋水仙堿、細胞松弛素B及丙烯酰胺分別調節(jié)軟骨細胞骨架的微管、肌動蛋白微絲及波形蛋白中間纖絲,從大體形態(tài)、組織切片染色、免疫組化染色及RT-PCR分析成軟骨細胞表型的改變。結果對照組、秋水仙堿組及細胞松弛素B組細胞較密集、均一,丙烯酰胺組細胞較少、分散。對照組、秋水仙堿組及細胞松弛素B組COLⅡ、蛋白多糖染色深,COLⅠ染色淺;而丙烯酰胺組COLⅡ、蛋白多糖染色淺,COLⅠ染色較深。RT-PCR數(shù)據(jù)統(tǒng)計分析顯示對照組與秋水仙堿組、細胞松弛素B組相比,COLⅠ(1.21±0.21 vs 1.55±0.06、1.70±0.07)及COLⅡ(1.10±0.12 vs 0.91±0.21、0.90±0.30)表達差異無統(tǒng)計學意義(P0.05);而對照組COLⅡ表達明顯多于丙烯酰胺組(0.26±0.06),COLⅠ表達明顯低于丙烯酰胺組(9.63±0.38),差異有統(tǒng)計學意義(P0.05)。結論丙烯酰胺調節(jié)軟骨細胞骨架波形蛋白中間纖絲可明顯下調COLⅡ、蛋白多糖的表達。
[Abstract]:Objective to study the relationship between the structural changes of chondrocytoskeleton protein and the phenotypic changes of chondroblasts. Methods the chondrocytes of knee joint of New Zealand rabbits were cultured by three-dimensional microgroup culture and the chondrocytes were divided into control group. In colchicine group, cytochalasin B group and acrylamide group, the microtubules, actin microfilaments and vimentin intermediate filaments of chondrocyte cytoskeleton were regulated by colchicine, cytochalasin B and acrylamide, respectively. The changes of chondroblast phenotype were analyzed from gross morphology, tissue section staining, immunohistochemical staining and RT-PCR.
