豐富環(huán)境對缺血缺氧腦損傷新生大鼠海馬腦源性神經(jīng)營養(yǎng)因子表達(dá)及細(xì)胞凋亡的影響

【摘要】:目的:探討豐富環(huán)境對缺血缺氧腦損傷(HIBD)新生大鼠海馬腦源性神經(jīng)營養(yǎng)因子(BDNF)表達(dá)及細(xì)胞凋亡的影響。方法:采用隨機(jī)數(shù)字表法將36只7日齡新生大鼠分為假手術(shù)組、模型組及豐富環(huán)境組,各組又分為術(shù)后7d、28d兩個亞組(n=6)。采用Rice-Vannucci經(jīng)典方法建立新生大鼠HIBD模型。模型組及假手術(shù)組不予任何干預(yù),豐富環(huán)境組予以早期撫觸及豐富環(huán)境刺激治療。術(shù)后7d、28d采用水迷宮檢測方法評估大鼠學(xué)習(xí)記憶能力,免疫組化法檢測大鼠海馬CA1區(qū)BDNF的表達(dá),TUNEL法檢測海馬CA1區(qū)細(xì)胞凋亡。結(jié)果:術(shù)后7d、28d,與模型組比較,豐富環(huán)境組逃避潛伏期均縮短(P0.05),穿越平臺次數(shù)增多(P0.05),BDNF表達(dá)升高(P0.05),TUNEL陽性細(xì)胞數(shù)降低(P0.05)。結(jié)論:豐富環(huán)境能改善HIBD新生大鼠學(xué)習(xí)記憶能力,其機(jī)制可能與上調(diào)海馬區(qū)BDNF的表達(dá)及抑制細(xì)胞凋亡有關(guān)。
