骨髓單個(gè)核細(xì)胞對(duì)腦梗死大鼠炎癥反應(yīng)的影響

【摘要】:目的探討骨髓單個(gè)核細(xì)胞(BMMNCs)是否通過抑制腦梗死大鼠腦內(nèi)的炎癥反應(yīng)來發(fā)揮神經(jīng)保護(hù)作用。方法選擇健康雄性SD大鼠,體質(zhì)量為250~300 g,線栓法制備永久性大腦中動(dòng)脈閉塞模型(p MCAO),造模成功后,隨機(jī)分為4組:假手術(shù)組(Sham組)、模型組(Model組)、溶劑組(Vehicle組)、骨髓單個(gè)核細(xì)胞組(BMMNCs組)。其中BMMNCs組于造模24 h后經(jīng)尾靜脈注射含1×107BMMNCs的PBS細(xì)胞懸液200μl;溶劑組注射等量的PBS溶液,分別于24 h、72 h、7 d采用Zea-Longa法進(jìn)行神經(jīng)功能評(píng)分;干濕重法檢測(cè)腦組織含水量;TTC法測(cè)定腦梗死體積;免疫組化、Western blot技術(shù)檢測(cè)炎性介質(zhì)環(huán)氧化酶-2(COX-2)、白介素-1β(IL-1β)的表達(dá)情況。結(jié)果 (1)各時(shí)間點(diǎn)模型組Zea-Longa評(píng)分、腦組織含水量和腦梗死體積均明顯高于假手術(shù)組(P0.05),BMMNCs組低于模型組,高于假手術(shù)組(P0.05),模型組與溶劑組相比無差異(P0.05);(2)各時(shí)間點(diǎn)模型組COX-2和IL-1β水平明顯均高于假手術(shù)組(P0.05),BMMNCs組低于模型組,高于假手術(shù)組(P0.05),模型組與溶劑組相比無差異(P0.05)。結(jié)論骨髓單個(gè)核細(xì)胞移植可通過抑制梗死后腦內(nèi)免疫炎癥反應(yīng),顯著改善腦梗死大鼠的神經(jīng)功能,這一治療作用可能與下調(diào)COX-2、IL-1β的表達(dá)有關(guān)。
[Abstract]:Objective to investigate whether bone marrow mononuclear cells (BMMNCs) play a neuroprotective role by inhibiting inflammatory response in the brain of rats with cerebral infarction. Methods healthy male SD rats were selected. The permanent middle cerebral artery occlusion (MCAO) model was established by the method of thread embolization. The rats were randomly divided into four groups: sham group, model group, vehicle group, bone marrow mononuclear cell group and bone marrow mononuclear cell group. The BMMNCs group was injected with PBS cell suspension containing 1 脳 107 BMMNCs through tail vein for 24 hours, and the solvent group was injected with the same amount of PBS solution. The neurological function was evaluated by Zea-Longa method at 24 hours and 72 hours after 7 days, and the volume of cerebral infarction was measured by dry and wet weight method. Immunohistochemistry Western blot technique was used to detect the expression of inflammatory mediator cyclooxygenase-2 (COX-2) and interleukin-1 尾 (IL-1 尾). Results: the Zea-Longa score, brain water content and cerebral infarction volume in model group were significantly higher than those in sham operation group (P 0.05) and the level of BMMNCs in model group was significantly higher than that in model group. The levels of COX-2 and IL-1 尾 in the model group were significantly higher than those in the sham operation group and the model group were significantly higher than those in the model group. There was no difference between the model group and the solvent group. Conclusion Bone marrow mononuclear cell transplantation can significantly improve the neural function of rats with cerebral infarction by inhibiting the immune inflammation after infarction. This therapeutic effect may be related to the down-regulation of the expression of COX-2 and IL-1 尾.
