【摘要】：目的實(shí)施大鼠體外循環(huán)(CPB)前后血漿比較蛋白質(zhì)組學(xué)研究,了解CPB前、后血漿蛋白質(zhì)的差異性表達(dá),以期發(fā)現(xiàn)對(duì)早期診斷CPB后并發(fā)癥有參考價(jià)值的血漿標(biāo)志物。方法將成年雄性SD大鼠10只分為實(shí)驗(yàn)組和對(duì)照組,每組5只,術(shù)前自由進(jìn)食進(jìn)水。實(shí)驗(yàn)組大鼠建立CPB模型,對(duì)照組只進(jìn)行麻醉和動(dòng)、靜脈穿刺操作。兩組動(dòng)物分別在CPB前和CPB結(jié)束時(shí)采取血液標(biāo)本1mL,肝素抗凝,分離血漿。血漿蛋白提純后進(jìn)行二維凝膠電泳、用ImageScanner掃描儀掃描成像。將被認(rèn)定為差異表達(dá)的蛋白質(zhì)斑點(diǎn)進(jìn)行切膠、酶解和肽段提取后,用質(zhì)譜儀分析鑒定。結(jié)果 CPB后,大鼠血漿蛋白質(zhì)表達(dá)斑點(diǎn)數(shù)目明顯增加,共有17個(gè)表達(dá)上調(diào)的蛋白質(zhì)斑點(diǎn)被認(rèn)定為CPB所致。經(jīng)質(zhì)譜分析,共鑒定出5種蛋白質(zhì),分別為:凝溶膠蛋白、結(jié)合珠蛋白、載脂蛋白A1(ApoA1)、免疫球蛋白和Ba1-647。結(jié)論 CPB可引起大鼠血漿蛋白質(zhì)出現(xiàn)差異性表達(dá),從功能上分析,凝溶膠蛋白、結(jié)合珠蛋白、ApoA1有可能作為研究CPB并發(fā)癥的血漿標(biāo)志物。
[Abstract]:Objective to investigate the differential expression of plasma protein before and after CPB in rats before and after cardiopulmonary bypass (CPB). Methods Ten adult male Sprague-Dawley rats were divided into experimental group and control group with 5 rats in each group. The CPB model was established in the experimental group and the control group only performed anesthesia and venipuncture. Before CPB and at the end of CPB, the two groups were treated with 1 mL of blood sample, heparin anticoagulant and plasma separation. The plasma protein was purified by two-dimensional gel electrophoresis and scanned by ImageScanner scanner. Protein spots identified as differentially expressed proteins were identified by mass spectrometer after gelatinization, enzymolysis and peptide extraction. Results after CPB, the number of protein expression spots in plasma of rats was significantly increased, and a total of 17 up-regulated protein spots were thought to be caused by CPB. Five kinds of proteins were identified by mass spectrometry, they were agglutinin, binding globin, apolipoprotein A1-ApoA1, immunoglobulin and Ba1-647. Conclusion CPB can induce differential expression of plasma protein in rats. In functional analysis, coagulation sol protein and bound globin ApoA1 may be used as plasma markers to study the complications of CPB.
