【摘要】：目的探討miR-16表達(dá)水平與金黃色葡萄球菌膿毒癥嚴(yán)重程度的關(guān)聯(lián)性,進(jìn)一步探討其潛在的臨床意義。方法收集金黃色葡萄球菌膿毒癥血液標(biāo)本共計(jì)32例,膿毒性休克、嚴(yán)重膿毒癥和一般膿毒癥各8例,不同年齡段的健康對(duì)照24例;另外,收集革蘭氏陰性菌膿毒癥血液標(biāo)本8例。用Trizol液裂解全血后提取microRNA,采用熒光定量PCR測(cè)定miR-16在不同組的表達(dá)水平(2-△△Ct法),用SPSS 13.0軟件分析各組之間的統(tǒng)計(jì)學(xué)差異。采用SPSS 13.0軟件將miR-16定量值與相應(yīng)CRP和PCT值進(jìn)行相關(guān)性分析。結(jié)果 miR-16表達(dá)水平與金黃色葡萄球菌膿毒癥嚴(yán)重程度呈明顯的負(fù)關(guān)聯(lián),各實(shí)驗(yàn)組與對(duì)照組相比均有統(tǒng)計(jì)學(xué)差異(P0.001),實(shí)驗(yàn)組之間也有統(tǒng)計(jì)學(xué)差異(P0.01)。miR-16表達(dá)水平與CRP和PCT值呈負(fù)相關(guān)性,相關(guān)系數(shù)分別為-0.561和-0.769。結(jié)論 miR-16表達(dá)與金黃色葡萄球菌膿毒癥有明顯的負(fù)關(guān)聯(lián)性,提示其可能作為該菌膿毒癥嚴(yán)重程度的標(biāo)記物。
[Abstract]:Objective to investigate the relationship between the expression of miR-16 and the severity of Staphylococcus aureus sepsis and its potential clinical significance.Methods A total of 32 blood samples of Staphylococcus aureus sepsis were collected, including 8 cases of septic shock, 8 cases of severe sepsis and 24 cases of normal sepsis.Blood samples of Gram-negative bacterial sepsis were collected from 8 cases.MicroRNAs were extracted from whole blood with Trizol solution. The expression level of miR-16 in different groups was determined by fluorescence quantitative PCR. The statistical difference was analyzed by SPSS 13.0 software.SPSS 13.0 software was used to analyze the correlation between the quantitative value of miR-16 and the corresponding CRP and PCT values.Results there was a significant negative correlation between the expression of miR-16 and the severity of Staphylococcus aureus sepsis. There was a significant difference between the experimental group and the control group (P 0.001). There was also a negative correlation between the expression of miR-16 and CRP and PCT between the experimental group and the control group.The correlation coefficients were -0.561 and -0.769, respectively.Conclusion there is a negative correlation between miR-16 expression and Staphylococcus aureus sepsis, which suggests that miR-16 may be a marker for the severity of sepsis of Staphylococcus aureus.
