缺氧誘導(dǎo)因子-1α對低氧誘導(dǎo)自噬的調(diào)控作用

【摘要】:目的通過體外觀察驗證缺氧誘導(dǎo)因子-1α(HIF-1α)對自噬相關(guān)分子的調(diào)控作用。方法采用Western blotting和免疫熒光技術(shù),檢測在常氧和缺氧培養(yǎng)體系下轉(zhuǎn)染質(zhì);蛴盟幬锔深A(yù)人上皮性卵巢癌細(xì)胞株后HIF-1α和自噬相關(guān)分子的表達變化。結(jié)果轉(zhuǎn)染HIF-1α質(zhì)粒后,在常氧狀態(tài)下過度表達HIF-1α,Beclin1亦見增加,LC3-Ⅰ轉(zhuǎn)換成LC3-Ⅱ。在低氧狀態(tài)下敲除HIF-1α后,Beclin 1表達可減少;原被低氧誘導(dǎo)的LC3-Ⅱ被抑制。喜樹堿類藥物NSC606985能抑制高轉(zhuǎn)移卵巢癌細(xì)胞株的HIF-1α自噬水平。結(jié)論低氧微環(huán)境能夠激活HIF-1α,并誘導(dǎo)上皮性卵巢細(xì)胞發(fā)生自噬。NSC606985在納摩爾濃度即可發(fā)揮對HIF-1α的調(diào)控作用,但可因細(xì)胞類型不同而有差異,呈現(xiàn)正負(fù)不同的調(diào)控。
