葡萄籽提取物減緩西雜肉?漳c上皮細(xì)胞熱應(yīng)激反應(yīng)的功能與機(jī)制研究

【摘要】:葡萄籽提取物(Grape seed extract,GSE)是多酚類(lèi)物質(zhì)的重要來(lái)源之一,具有抗氧化、抗炎和免疫調(diào)節(jié)的功能。本試驗(yàn)旨在研究GSE緩解肉?漳c上皮細(xì)胞(Jejunum epithelial cells,JEC)熱應(yīng)激反應(yīng)的作用與機(jī)制。在肉牛(西門(mén)塔爾♂×秦川牛Q的雜交后代)空腸上皮細(xì)胞培養(yǎng)基中分別添加0、10、20、40和80μg/mL的GSE,然后將各濃度梯度下的JEC細(xì)胞分別在37℃、40℃、43℃和46℃下進(jìn)行2h的培養(yǎng)。研究顯示,當(dāng)細(xì)胞培養(yǎng)溫度為40℃時(shí),添加GSE時(shí)細(xì)胞谷胱甘肽過(guò)氧化物酶(GSH-Px)活性(P=0.04)、細(xì)胞活力(P=0.004)、細(xì)胞表皮生長(zhǎng)因子(Epidermal growth factor,EGF;P=0.03)和細(xì)胞表皮生長(zhǎng)因于受體(Epidermal growth factor receptor,EGFR;P=0.01)mRNA表達(dá)均有升高。在同一溫度下,添加GSE降低細(xì)胞中自細(xì)胞介素(Interleukin 8.IL-8)(P=0.01)、腫瘤壞死因子α(TNF-α)mRNA的表達(dá),同時(shí)降低IL-1β(P=0.02)、Toll樣受體4(TLR4;P=0.04)蛋白和熱休克蛋白70(HSP70;P0.001)的濃度。當(dāng)細(xì)胞培養(yǎng)溫度為43℃時(shí),添加GSE組細(xì)胞的過(guò)氧化氫酶活性(P=0.04)、細(xì)胞活力(P0.001).EGF(P0.001)和EGFR(P0.001)mRNA的表達(dá)均得到提升,而IL-1β(P0.001).TLR4(P=0.03)和HSP70(P0.001)的濃度以及IL-8(P0.001)、TLR4(P=0.002)和TNF-α(P0.001)的mRNA表達(dá)卻顯著降低。T×GSE互作效應(yīng)在JEC細(xì)胞IL-1β(P0.001)、IL-8(P0.001)、TNF-α(P=0.01)、HSP70(P=0.04)的濃度和細(xì)胞活力(P0.001)上得到體現(xiàn)。本試驗(yàn)研究結(jié)果表明,在特定溫度范圍內(nèi),GSE能夠從提高細(xì)胞抗氧化能力以及降低炎性反應(yīng)兩個(gè)方面緩解JEC細(xì)胞的熱應(yīng)激反應(yīng)。
