松遼黑豬四種基因多態(tài)性與背膘厚的關聯(lián)分析

【摘要】:背膘厚是家豬重要的經濟性狀,同時也是地方豬種選育過程中主要的育種指標。為深入研究松遼黑豬群體背膘厚的遺傳基礎,以173頭松遼黑豬背最長肌組織、四點背膘處的脂肪組織為試驗樣本,對MC4R、HDAC1、LEP及OLR1進行基因多態(tài)性檢測,進而分析其與背膘厚、肌內脂肪含量間的關聯(lián)性,以期為降低背膘厚、提高瘦肉率提供理論基礎及分子標記。本研究結果如下:飼養(yǎng)差異:放牧組的胸腰結合處背膘厚、腰薦結合處背膘厚及平均背膘厚顯著低于舍飼組。性狀分析:肌內脂肪含量與肩部最厚處背膘厚間存在強相關,與平均背膘厚間存在顯著相關;體重、肩部最厚、六七肋、胸腰結合、腰薦結合處背膘厚及平均背膘厚間均存在強相關。多態(tài)性:MC4R、HDAC1、LEP及OLR1四個候選基因在松遼黑豬群體中均發(fā)現(xiàn)多態(tài)位點,且均處于Hardy-Weinberg平衡狀態(tài),遺傳多態(tài)性均為中度多態(tài),遺傳變異程度均處于中等水平。經分析發(fā)現(xiàn),HDAC1基因10806位點處GA多態(tài)性與六七肋、腰薦結合處、最厚肋處的背膘厚密切相關。OLR1基因9011位點處TC、9012位點處CA均與肌內脂肪間存在關聯(lián)。9070位點處GA多態(tài)性與四點背膘厚間存在顯著聯(lián)系。MC4R基因、LEP基因在松遼黑豬群體中與各點背膘厚、肌內脂肪含量間未顯示存在任何相關性。表達量:HDAC1基因高背膘厚組在六七肋、胸腰結合處脂肪組織中的mRNA表達量顯著高于低背膘厚組(P0.05),OLR1基因高背膘厚組在四點背膘組織中的mRNA表達量均高于低背膘厚組且差異顯著(P0.05)。MC4R、LEP基因兩組在四點處脂肪組織中的mRNA表達量間均無顯著性差異。
[Abstract]:Backfat thickness is an important economic trait in domestic pigs, and it is also the main breeding index in the breeding process of local pigs. In order to study the genetic basis of backfat thickness in Songliao black pig population, the genetic polymorphisms of MC4RN HDAC1LEP and OLR1 were detected by using the longissimus dorsi muscle tissue of 173 Songliao black pigs and adipose tissue at four-point backfat, and the genetic polymorphisms of MC4RHDAC1LEP and OLR1 were analyzed. The correlation of intramuscular fat content in order to provide theoretical basis and molecular marker for reducing backfat thickness and increasing lean meat percentage. The results were as follows: the feeding difference: the backfat thickness of thoracolumbar joint, backfat thickness and average backfat thickness of the grazing group were significantly lower than those of the house-fed group. Character analysis: there was strong correlation between intramuscular fat content and backfat thickness at the thickest part of shoulder, and significant correlation between intramuscular fat content and average backfat thickness. There was strong correlation between backfat thickness and average backfat thickness. Polymorphic loci were found in four candidate genes of HDAC1LEP and OLR1 in Songliao Black Pig population, and they were all in Hardy-Weinberg equilibrium. The genetic polymorphisms were moderately polymorphic, and the degree of genetic variation was at the medium level. The genetic polymorphism at 10806 locus of HDAC1 gene was found to be associated with the combination of six or seven ribs and waist recommendation. The backfat thickness of the thickest rib was closely related to the backfat thickness of the most thick rib. There was a significant association between the 9011 locus of OLR1 gene and the TCQ9012 locus CA. There was a significant association between the polymorphism of GA at the locus of .9070 and the backfat thickness of the four-point backfat. There was a significant association between the backfat thickness and the backfat thickness at each point in the Songliao Black Pig population. There was no correlation between intramuscular fat content. HDAC1 gene was expressed in HDAC1 group with high backfat thickness. The mRNA expression level in thoracolumbar binding adipose tissue was significantly higher than that in low backfat thickness group (P0.05). The mRNA expression of OLR1 gene in high backfat thickness group was higher than that in low backfat thickness group and the difference was significant (P0.05). MC4RNLEP gene expression level was significantly higher in four-point fat group than in low-backfat thickness group (P0.05). There was no significant difference in mRNA expression between tissues.
