【摘要】:抗菌肽(antibacterial peptides,ABP)是生物體內(nèi)產(chǎn)生的一種具有廣譜抗菌作用的小分子多肽,屬非特異性防疫系統(tǒng)的重要組份。人表皮生長(zhǎng)因子(hEGF)作為促生長(zhǎng)因子家族成員之一,具有促進(jìn)新血管形成和表皮細(xì)胞增殖的作用,是上皮細(xì)胞、表皮細(xì)胞和間充質(zhì)等敏感細(xì)胞的絲裂原,具有促進(jìn)生長(zhǎng)和提高免疫的作用。抗菌肽AWRK6是經(jīng)Dybowskin-2CDYa設(shè)計(jì)改造而得的新型抗菌肽,與以往發(fā)現(xiàn)的其它蛙類抗菌肽相比相似性小,它富含賴氨酸,具有親水性強(qiáng),穩(wěn)定性高,抑菌活性強(qiáng)和抑菌譜廣等特點(diǎn)。hEGF-AWRK6是由本實(shí)驗(yàn)室將人表皮生長(zhǎng)因子hEGF和抗菌肽AWRK6的基因DNA連接、轉(zhuǎn)化并表達(dá)純化得到的重組融合多肽。本實(shí)驗(yàn)以肉雞為實(shí)驗(yàn)動(dòng)物,分析了融合肽hEGF-AWRK6對(duì)肉雞生產(chǎn)性能、養(yǎng)分代謝、抗氧化和免疫機(jī)能的影響。探討了融合肽hEGF-AWRK6替代飼用抗生素應(yīng)用于家畜養(yǎng)殖的可行性。實(shí)驗(yàn)選取84只1日齡健康A(chǔ)A肉仔雞,隨機(jī)分4組。對(duì)照組飼喂基礎(chǔ)日糧,實(shí)驗(yàn)Ⅰ組、Ⅱ組和抗生素組分別在基礎(chǔ)日糧中添加50mg/kg、100mg/kg的融合肽hEGF-AWRK6和5mg/kg的黃霉素,實(shí)驗(yàn)周期為42d。結(jié)果顯示:1、與對(duì)照組相比,實(shí)驗(yàn)Ⅱ組與抗生素組的肉雞體重分別提高了21.4%(p0.01)和19.0%(p0.05),半凈膛、全凈膛和胸肌重亦顯著增加,且實(shí)驗(yàn)Ⅱ組優(yōu)于抗生素組,而各組肉雞的全凈膛率、半凈膛率以及腿肌重沒有顯著差異;2、實(shí)驗(yàn)Ⅱ組肉雞免疫器官指數(shù)、脾臟和巨噬細(xì)胞細(xì)胞活力及Sig-A水平,與對(duì)照組相比分別提高了66.67%,27.27%,68.29%和54.11%,差異顯著,且實(shí)驗(yàn)Ⅱ組優(yōu)于抗生素組。3、與對(duì)照組相比,實(shí)驗(yàn)Ⅱ組的肉雞肝臟的GSH-PX、T-AOC和SOD水平分別提高了的23.07%、64.6%、10.81%,并且降低MDA水平的28.57%,差異顯著。4、融合肽hEGF-AWRK6能夠顯著提高肉雞肌肉蛋白質(zhì)和脂肪得含量,與對(duì)照組相比,實(shí)驗(yàn)Ⅱ組的肌肉蛋白質(zhì)和脂肪分別提高了15.89%(p0.01)和29.31%(p0.01)。此外,融合肽hEGF-AWRK6可提高肉雞小腸絨毛高度和隱窩深度比值,降低肉雞腸道中有害菌群大腸桿菌的數(shù)量。本文研究結(jié)論:在日料糧中添加100mg/kghEGF-AWRK6可以顯著增加肉雞的日增重,提高飼料的轉(zhuǎn)化效率,提高肉雞肌肉中蛋白和脂肪的含量;融合肽hEGF-AWRK6可提高肉雞血清中的免疫球蛋白A(Sig-A)水平,并能顯著提高肉雞的肝臟抗氧化機(jī)能,增強(qiáng)肉雞的抵抗力;融合肽hEGF-AWRK6可改善肉雞腸道菌群和腸道結(jié)構(gòu),降低腸道中的大腸桿菌數(shù)量;日料中添加高劑量hEGF-AWRK6(100mg/kg)組的肉雞的生產(chǎn)性能及免疫能力優(yōu)于日料中添加抗生素組的肉雞。本實(shí)驗(yàn)為hEGF-AWRK6作為飼用抗生素替代品在家畜飼養(yǎng)中的應(yīng)用提供了實(shí)驗(yàn)依據(jù)。
[Abstract]:Antimicrobial peptide (ABP) is a kind of small molecular peptide with broad-spectrum antimicrobial activity produced in vivo, which is an important component of nonspecific epidemic prevention system. Human epidermal growth factor hEGF, as a member of the growth promoting factor family, is a mitogen of epithelial cells, epidermal cells and mesenchymal cells, which can promote the formation of new blood vessels and the proliferation of epidermal cells. It has the function of promoting growth and improving immunity. Antimicrobial peptide AWRK6 is a new antimicrobial peptide designed and modified by Dybowskin-2CDYa. It has less similarity with other frog antimicrobial peptides found in the past. It is rich in lysine, has strong hydrophilicity and high stability. HEGF-AWRK6 is a recombinant fusion polypeptide which is expressed and expressed by the DNA of human epidermal growth factor (hEGF) and antimicrobial peptide AWRK6 (AWRK6), which is characterized by strong bacteriostatic activity and wide antimicrobial spectrum. The effects of fusion peptide hEGF-AWRK6 on performance, nutrient metabolism, antioxidation and immune function of broilers were studied.
