腺嘌呤制備陽(yáng)虛型慢性腎功能衰竭大鼠模型綜合指標(biāo)及其機(jī)理研究

【摘要】:研究目的:本課題通過動(dòng)物實(shí)驗(yàn),對(duì)相關(guān)檢驗(yàn)指標(biāo)研究,開發(fā)適用于防治陽(yáng)虛型慢性腎功能衰竭中醫(yī)藥藥理學(xué)研究實(shí)用、穩(wěn)定的動(dòng)物模型,,探討陽(yáng)虛型慢性腎功能衰竭SD大鼠模型的客觀化指標(biāo)、病理機(jī)制及其與人類CRF病變的異同。 材料與方法:采用健康SD大鼠24只,正常飼養(yǎng)7天后,隨機(jī)分為腺嘌呤灌胃模型組(12只)、空白對(duì)照組(12只)。腺嘌呤灌胃模型組:按200mg/kg·d劑量將腺嘌呤制成混懸液2ml,隔日1次,定時(shí)經(jīng)灌胃給藥,累計(jì)總劑量為3g/只,全過程30d;空白對(duì)照組:正常飼料喂養(yǎng),全過程30d。全部動(dòng)物均自由攝取飼料及飲水。兩組動(dòng)物在第31天采用常規(guī)麻醉后處死,經(jīng)腹主動(dòng)脈取血10ml做相關(guān)實(shí)驗(yàn)室檢查,取雙腎按光鏡、電鏡病理學(xué)要求取樣送檢。 選擇腎臟病理學(xué)、血液生化(腎功、肝功、電解質(zhì))、一氧化氮(NO)、內(nèi)皮素(ET)、環(huán)磷酸鳥苷(cGMP)、環(huán)磷酸腺苷(cAMP)、IgG、IgM、血栓素(TXB2)、前列腺素(6-K—PGFla)、腫瘤壞死因子(TNF-a)、白介素(IL-1β)及中分子物質(zhì)(MMS)等客觀指標(biāo)進(jìn)行研究,統(tǒng)計(jì)分析各項(xiàng)指標(biāo)在實(shí)驗(yàn)中的變化及其意義,綜合闡述腺嘌呤制備陽(yáng)虛型慢性腎功能衰竭動(dòng)物模型的發(fā)病機(jī)理,及其與人類慢性腎功能衰竭病理改變的異同。 結(jié)果: 1 臨床表現(xiàn):模型組SD大鼠在用藥12-15d開始出現(xiàn)多尿、精神萎靡、眼瞼周圍浮腫、體重增長(zhǎng)緩慢等一系列陽(yáng)虛型腎功能衰竭臨床表現(xiàn),空白組外觀及活動(dòng)正常,體重上升。 2 模型組SD大鼠腎功能檢測(cè)與空白對(duì)照組比較差異性顯著(P<0.01),但尿酸含量空白組大于模型組; 3 模型組SD大鼠電解質(zhì)檢測(cè)與空白對(duì)照組比較,鉀、磷差異性顯著(P<0.01),鈉、鈣、氯化物均無明顯差異(P>0.05);
[Abstract]:Objective: to develop a practical and stable animal model of TCM pharmacology for the prevention and treatment of chronic renal failure due to Yang deficiency through animal experiments. Objective: to explore the objective indexes, pathological mechanism and the differences and similarities between SD rat model of chronic renal failure with yang deficiency and human CRF. Materials and methods: 24 healthy SD rats were randomly divided into adenine perfusion model group (n = 12) and blank control group (n = 12) after 7 days of normal feeding. In the model group, adenine was made into suspension at the dose of 200mg/kg d, once every other day. The cumulative total dose of adenine was 3g/ only for 30 days, while the blank control group was fed with normal diet for 30 days. All animals are free to eat feed and drinking water. The animals of the two groups were killed after routine anesthesia on the 31st day. Blood 10ml was taken from the abdominal aorta for laboratory examination. The two kidneys were taken for sampling according to light microscope and electron microscope pathology. Choose renal pathology, blood biochemistry (renal function, liver function), Electrolyte, nitric oxide (no), endothelin (et), cyclic guanosine monophosphate (cGMP), cyclic adenosine monophosphate (camp) IgGG, thromboxane (TXB2), prostaglandin (6-K-PGFla), tumor necrosis factor (TNF-a), interleukin-1 尾 (IL-1 尾) and mesomolecular substance (MMS) were studied. The changes of various indexes in the experiment and their significance were analyzed. The pathogenesis of the animal model of chronic renal failure caused by adenine deficiency and the similarities and differences between the pathological changes of chronic renal failure and that of human chronic renal failure were expounded. Results: 1 Clinical manifestations: SD rats in the model group began to develop polyuria, mental depression and edema around the eyelid at 12-15 days after treatment. A series of clinical manifestations of renal failure of Yang deficiency type, such as slow weight gain, normal appearance and activity in blank group, 2 the renal function of SD rats in the model group was significantly different from that in the blank control group (P < 0.01), but the uric acid content in the blank group was higher than that in the model group. (3) compared with the control group, the electrolyte of SD rats in the model group showed significant difference in potassium and phosphorus (P < 0.01), but there was no significant difference in sodium, calcium and chloride (P > 0.05).
