【摘要】：豚鼠氣單胞菌(Aeromonas caviae)是養(yǎng)殖及野生魚、蝦、蛙、甲魚等生物常見的細菌性病原體，可通過受損傷的皮膚及傷口感染上述生物，，并可進一步通過食物鏈感染人。此菌所致魚類等豚鼠氣單胞菌病具有流行廣、發(fā)病快、死亡率高等特點，常常造成養(yǎng)殖漁業(yè)的巨大經(jīng)濟損失，嚴重危害漁業(yè)及其它水產(chǎn)業(yè)的發(fā)展。目前國內(nèi)外有關(guān)豚鼠氣單胞菌魚病的研究多局限于病原微生物學方面，對該病的診斷主要依靠臨診病理觀察及細菌學鑒定，兩種方法在準確性及檢測速度上均有一定限制；防治方面主要利用抗生素或減活、滅活疫苗，安全性和有效性均不理想。鑒于此，本研究設計制備抗豚鼠氣單胞菌單克隆抗體并進而制備其獨特型單克隆抗體，為豚鼠氣單胞菌病的檢測及防治提供新的技術(shù)方法，并為其它魚用疫苗的研制提供參考。具體實驗方法及結(jié)果如下： (1) 通過動物免疫、細胞融合、篩選、克隆化及利用ELISA檢測技術(shù)，制備并鑒定抗豚鼠氣單胞菌的單克隆抗體(mAb)。共得到7株能穩(wěn)定分泌mAb的細胞株，分別命名為：0E10、1A2、1C4、1F1、1F10、1H8和2A1。染色體鑒定結(jié)果7株細胞染色體均符合雜交瘤細胞染色
[Abstract]:Aeromonas guinea pig (Aeromonas caviae) is a common bacterial pathogen of aquaculture and wild fish, shrimp, frog, turtle and other organisms. It can infect the above organisms through the injured skin and wound, and can further infect people through the food chain. Aeromonas aeruginosa, such as fishes and other guinea pigs, is characterized by widespread epidemic, fast onset and high mortality. It often causes huge economic losses in aquaculture, and seriously endangering the development of fishery and other aquatic industries. At present, the domestic and foreign research on Aeromonas caviae disease confined to the microbiology, diagnosis of the disease mainly depends on clinical pathology and bacteriology, two kinds of methods in both accuracy and speed of detection is limited; prevention mainly using antibiotics or live attenuated and inactivated vaccine, safety and the effectiveness is not ideal. In view of this, we designed and prepared monoclonal antibodies against Aeromonas guinea pigs, and then prepared their unique monoclonal antibodies, providing new technical methods for the detection and prevention of Aeromonas guinea pigs, and providing references for the development of other fish vaccines.
