營(yíng)養(yǎng)限制與補(bǔ)償對(duì)蒙古羔羊血常規(guī)和瘤胃及盲腸生長(zhǎng)發(fā)育影響

【摘要】:本試驗(yàn)研究營(yíng)養(yǎng)限制與補(bǔ)償對(duì)蒙古羔羊血常規(guī)和瘤胃及盲腸生長(zhǎng)發(fā)育影響,試驗(yàn)選用30只體況正常,平均體重為20.6 kg的4月齡蒙古羔羊。隨機(jī)分為對(duì)照組(CG)、限制組(RG),營(yíng)養(yǎng)限制期(60 d)結(jié)束后,隨機(jī)在各組挑選5只進(jìn)行屠宰,每組剩余10只羊,進(jìn)入補(bǔ)償期飼喂,各分成兩組,每組各五只羊,CH、RH組飼喂精粗比為5:5的粗飼料為青干草的日糧,CM、RM組飼喂精粗比為7:3粗飼料為秸稈的高精料日糧。1.營(yíng)養(yǎng)限制與補(bǔ)償對(duì)蒙古羔羊血液生理指標(biāo)的影響。分別在試驗(yàn)的60 d、90 d、120 d通過(guò)對(duì)羔羊頸靜脈采血,應(yīng)用血常規(guī)自動(dòng)分析儀進(jìn)行血常規(guī)四類十二項(xiàng)指標(biāo)測(cè)試,對(duì)數(shù)據(jù)結(jié)果進(jìn)行分析。結(jié)果顯示:營(yíng)養(yǎng)限制結(jié)束,R組WBC顯著低于C組(P0.01),其他各項(xiàng)指標(biāo)均有下降趨勢(shì);與對(duì)照組(CH、CM)相比,補(bǔ)償30d,限制各組PLT、WBC均有顯著性升高;補(bǔ)償60d,限制各組HGB均呈現(xiàn)上升趨勢(shì)。整體補(bǔ)償階段限制各組RBC均呈下降趨勢(shì)。2.營(yíng)養(yǎng)限制與補(bǔ)償對(duì)蒙古羔羊瘤胃、盲腸形態(tài)學(xué)影響通過(guò)HE染色、顯微鏡等技術(shù),對(duì)瘤胃、盲腸黏膜進(jìn)行觀察,研究營(yíng)養(yǎng)限制及補(bǔ)償,對(duì)瘤胃、盲腸黏膜上皮形態(tài)結(jié)構(gòu)的影響。結(jié)果顯示:營(yíng)養(yǎng)限制結(jié)束,限制組瘤胃乳頭長(zhǎng)度顯著縮短(P0.01),角質(zhì)層厚度變薄;盲腸黏膜厚度顯著降低(P0.01),杯狀細(xì)胞顯著減少(P0.01),嗜伊紅細(xì)胞、球狀白細(xì)胞顯著增多(P0.05;P0.01)。補(bǔ)償后,RH、RM組瘤胃乳頭高度及角質(zhì)層厚度有所上升(P0.05),乳頭寬度呈現(xiàn)下降趨勢(shì);盲腸RM組嗜伊紅細(xì)胞顯著增多(P0.01),RH組球狀白細(xì)胞顯著增多(P0.01),其余各項(xiàng)指標(biāo)無(wú)顯著變化。3.營(yíng)養(yǎng)限制與補(bǔ)償對(duì)蒙古羔羊瘤胃、盲腸細(xì)胞增殖增肥與凋亡的影響利用紫外分光光度計(jì)及酶標(biāo)儀,對(duì)瘤胃、盲腸的總DNA含量和總蛋白含量進(jìn)行測(cè)定。結(jié)果顯示:營(yíng)養(yǎng)限制結(jié)束,瘤胃、盲腸細(xì)胞數(shù)有所增加(P0.05)。補(bǔ)償后,瘤胃與盲腸呈現(xiàn)不同的變化:限制組與對(duì)照組相比,瘤胃細(xì)胞數(shù)量增多;盲腸細(xì)胞則與限制期數(shù)量持平。采用TUNEL法并用顯微鏡觀察試驗(yàn)各階段瘤胃和盲腸上皮細(xì)胞的細(xì)胞凋亡。結(jié)果顯示:營(yíng)養(yǎng)限制結(jié)束,瘤胃、盲腸發(fā)現(xiàn)細(xì)胞凋亡數(shù)量顯著增多(P0.01);補(bǔ)償后,瘤胃、盲腸細(xì)胞凋亡率均恢復(fù)到正常水平(P0.05)。4.營(yíng)養(yǎng)限制與補(bǔ)償對(duì)蒙古羔羊瘤胃、盲腸組織細(xì)胞連接相關(guān)蛋白基因的影響通過(guò)實(shí)時(shí)熒光定量分析小窩蛋白、間隙連接蛋白、緊密連接蛋白,在瘤胃、盲腸內(nèi)mRNA的表達(dá)量。結(jié)果顯示:胃腸道內(nèi)環(huán)境的差異,營(yíng)養(yǎng)限制階段,限制組在盲腸組織的表達(dá)量高于瘤胃組織的表達(dá)量,且高于對(duì)照組。補(bǔ)償階段,兩種補(bǔ)償方式對(duì)瘤胃、盲腸組織各基因的影響無(wú)較大差異。
[Abstract]:The effects of nutritional restriction and compensation on blood routine test and growth and development of rumen and cecum of Mongolian lambs were studied. Mongolian lamb, with an average weight of 20.6 kg, was randomly divided into two groups: control group (CGG), restriction group (RGG), nutrition restriction period (60 days). 5 sheep were slaughtered at random in each group. The remaining 10 sheep in each group were fed into the compensatory period and were divided into two groups. Five sheep in each group were fed with high concentrate diet of 7: 3 and straw. 1. The effect of nutrition limitation and compensation on blood physiological index of Mongolian lamb was studied. The blood samples were collected from the jugular vein of the lamb at 60 days, 90 days and 120 days, respectively. 12 items of blood routine were tested by automatic blood routine analyzer, and the data were analyzed. The results showed that the WBC of group R was significantly lower than that of group C (P 0.01), and the other indexes had a downward trend. After 30 days of compensation, there was a significant increase of RBC in each group, and 60 days of compensation, the HGB of all groups showed an increasing trend. The RBC of all groups was decreased in the whole compensation stage. 2. Nutrition restriction and compensation were used in the rumen of Mongolian lamb. The morphological effects of the caecum were observed by HE staining and microscope. The effects of nutritional limitation and compensation on the morphology and structure of the epithelium of rumen and caecum were studied. In the restricted group, the length of rumen papilla was shortened significantly, the thickness of cuticle became thinner, the thickness of caecum mucosa decreased significantly, and the goblet cells significantly decreased P0.01, eosinophilic erythrocyte. The height of rumen papilla and the thickness of cuticular layer in RHNRM group increased after compensation, and the width of nipple decreased. The eosinophilic erythrocytes in the cecum RM group were significantly increased and the globular white blood cells were increased significantly in the RH group. The other indexes had no significant change. 3. Nutritional restriction and compensation on the rumen of Mongolian lamb. Effects of Cecum Cell Proliferation, Fertilizer and apoptosis the contents of total DNA and total protein in rumen and caecum were measured by ultraviolet spectrophotometer and enzyme marker. The number of cecum cells increased (P0.05A). After compensation, the rumen and caecum showed different changes: compared with the control group, the number of rumen cells in the restricted group increased; The number of cecum cells was the same as that of limiting period. The apoptosis of rumen and cecum epithelial cells was observed by TUNEL method and microscope. The results showed that the end of nutrition restriction, the end of rumen, The number of apoptosis in the caecum was significantly increased (P 0.01), and the apoptotic rate of rumen and caecum cells returned to the normal level after compensation. The expression of mRNA in rumen and caecum by real-time fluorescence quantitative analysis of fossa protein, gap junction protein and tight junction protein. In nutritional restriction stage, the expression of cecum in the restriction group was higher than that in the rumen tissue and higher than that in the control group. In the compensatory stage, there was no significant difference in the effect of the two compensation modes on the genes in rumen and caecum tissues.
