第一部分血漿外泌體Cripto-1在胃癌診斷和預后判斷中的應用價值分析目的:胃癌（astric cancer,GC）是一種常見的惡性腫瘤,由于發(fā)現(xiàn)較晚、具有侵襲性、治療反應差導致胃癌預后較差,嚴重危害人們的生命健康。外泌體通過傳遞蛋白質(zhì)、RNA和DNA等功能分子參與細胞間的通訊,在腫瘤液體活檢中具有廣闊的應用前景。然而,目前Cripto-1或人畸胎瘤衍生生長因子1（tdgf1）在循環(huán)外泌體中的表達情況及外泌體Cripto-1在GC中的臨床價值尚無相關(guān)報道。為此,本課題探究外泌體Cripto-1在GC患者血漿中的表達及其在GC診斷及預后判斷中的價值。方法:1.運用超速離心法提取GC細胞系SGC7901和BGC823培養(yǎng)上清中外泌體和血漿外泌體,通過電鏡觀察、粒徑分析和western blot實驗進行鑒定。Western blot實驗觀察Cripto-l在GC細胞系及釋放外泌體中的表達情況。2.建立exo-ELISA方法,檢測118例GC患者、62例非萎縮性胃炎、26例胃息肉、62例萎縮性胃炎和60例健康對照者血漿外泌體Cripto-1表達水平。3.采用受試者工作特征（ROC）曲線評價血漿...
