采用高壓均質(zhì)法制備阿苯達(dá)唑納米脂質(zhì)體的工藝優(yōu)化

【摘要】:采用單因素和Box-Behnken設(shè)計(jì)-效應(yīng)面法優(yōu)化了微射流高壓均質(zhì)法制備阿苯達(dá)唑納米脂質(zhì)體的處方和工藝參數(shù)。分別以均質(zhì)壓力、均質(zhì)溫度、阿苯達(dá)唑與大豆磷脂質(zhì)量比為考察對(duì)象,以基于平均粒徑和包封率的綜合指標(biāo)為評(píng)價(jià)指標(biāo),在單因素考察結(jié)果的基礎(chǔ)上采用3因素3水平星點(diǎn)設(shè)計(jì)繼續(xù)試驗(yàn),對(duì)結(jié)果進(jìn)行二項(xiàng)式擬合,再用效應(yīng)面法篩選出最佳工藝條件。結(jié)果確定最優(yōu)工藝為:制備溫度37℃,均質(zhì)壓力137.90 MPa,均質(zhì)次數(shù)8次,阿苯達(dá)唑與大豆磷脂質(zhì)量比為1∶2。所得納米脂質(zhì)體的粒徑為(208.25±2.13)nm,包封率為(96.16±0.74)%,粒徑與包封率的實(shí)測(cè)值與預(yù)測(cè)值的偏差分別為0.94%和0.05%。透射電鏡顯示納米脂質(zhì)體粒徑均一,呈球狀分布;在室溫及4℃放置6個(gè)月穩(wěn)定性良好。
[Abstract]:The formulation and technological parameters of albendazole nanoliposomes were optimized by single factor and Box-Behnken design-effect surface method.The homogenization pressure, homogenization temperature, mass ratio of albendazole to soybean phospholipid were investigated, and the comprehensive indexes based on average particle size and entrapment efficiency were used as the evaluation index.On the basis of the results of single factor investigation, three factors and three levels of star point design were used to continue the experiment, and the binomial fitting was carried out, and the optimum technological conditions were selected by using the effect surface method.The results showed that the optimum conditions were as follows: preparation temperature 37 鈩,
