【摘要】：目的:比較3種堿性染液對(duì)顯微捕獲聯(lián)合低體積擴(kuò)增(Lowvolume polymerase chain reaction,LVPCR)技術(shù)的影響,為細(xì)胞分離檢驗(yàn)中細(xì)胞核標(biāo)記物的選擇提供依據(jù)。方法:顯微捕獲聯(lián)合LVPCR中,用蘇木素、龍膽紫、亞甲基藍(lán)3種染液對(duì)口腔上皮細(xì)胞進(jìn)行標(biāo)識(shí)。參照龍膽紫染色法,比較0.5、1、2、5、8μl蘇木素染液及2、5、8、10、15μl亞甲基藍(lán)染液的染色效果,優(yōu)化染色條件;并比較優(yōu)化后的細(xì)胞核標(biāo)識(shí)效果。顯微操作儀捕獲3種染液染色后的細(xì)胞進(jìn)行DNA檢驗(yàn),比較檢測(cè)結(jié)果;并捕獲染色10、30、60 min口腔上皮細(xì)胞,比較染色時(shí)間對(duì)DNA檢測(cè)結(jié)果的影響。結(jié)果:100μl口腔上皮細(xì)胞懸液加入2μl蘇木素、0.5μl龍膽紫或5μl亞甲基藍(lán)染液,細(xì)胞核均著色明顯,與胞漿對(duì)比清晰。5個(gè)細(xì)胞的分離檢驗(yàn)結(jié)果示有效分型率分別為90%、100%、66.7%,成功分型率為20%、100%、11.1%;龍膽紫、蘇木素染色時(shí)間(≤60 min)對(duì)DNA檢測(cè)結(jié)果無(wú)明顯影響,而亞甲基藍(lán)染色時(shí)間延長(zhǎng)導(dǎo)致等位基因丟失等現(xiàn)象加重,影響檢測(cè)結(jié)果。結(jié)論:3種染液均可清晰標(biāo)識(shí)細(xì)胞核;而龍膽紫染液對(duì)LVPCR技術(shù)抑制最弱,為三者中較優(yōu)的細(xì)胞核標(biāo)記物。
[Abstract]:Objective: To compare the 3 kinds of alkaline solution of microdissection combined with low volume amplification (Lowvolume polymerase chain reaction, LVPCR) the impact of technology, provide the basis for cell separation and cell nucleus marker selection in the test. Methods: microdissection combined with hematoxylin eosin, LVPCR, gentian violet, methylene blue dye to identify 3 kinds of oral epithelial cells. According to crystal violet staining method, dyeing effect of the 0.5,1,2,5,8 L and 2,5,8,10,15 l hematoxylin dye methylene blue dye, optimal dyeing conditions; cell identification results and to compare the optimized micro operation instrument. Capturing 3 staining of cells by using DNA test, comparison of test results and capture 10,30,60 min staining; oral epithelial cells, staining time impact on the results of DNA. Results: 100 L oral epithelial cell suspension into 2 l hematoxylin, 0.5 L or 5 L methylene blue gentian violet liquid, The nuclei were stained in cytoplasm and obvious contrast result in clear separation and inspection.5 cells showed effective typing rate were 90%, 100%, 66.7%, classification success rate was 20%, 100%, 11.1%; longdanzi hematoxylin staining time (less than 60 min) had no obvious effect on the detection results of DNA and methylene blue prolonged increase allele loss, affect the detection results. Conclusion: the 3 kinds of dye can be clearly identified and gentian violet stain nuclei; weakest inhibition on LVPCR technology, nuclear marker is the best in the three.
