心臟成纖維細胞旁分泌能力影響小鼠胚胎干細胞的命運(英文)

本研究旨在探討心臟成纖維細胞旁分泌能力對小鼠胚胎干細胞的影響。新生小鼠和成年小鼠成纖維細胞的條件培養(yǎng)基(ConM-NCF、ConM-ACF)分別稀釋50倍(1:50)或5倍(1:5),通過RT-real time PCR技術、ELISA、細胞增殖檢測以及計數(shù)心肌分化過程中跳動擬胚體(embryoid body, EB)的比例,觀察這些條件培養(yǎng)基對小鼠胚胎干細胞的影響。結果顯示,成纖維細胞細胞因子的分泌能力在發(fā)育過程中發(fā)生顯著變化,其中白細胞介素6 (interleukin 6, IL6)隨發(fā)育成熟顯著增加。ConM-NCF抑制干細胞的增殖,但高、低濃度對干性基因表達的作用不同。Con M-ACF 1:50促進干細胞增殖并上調干性基因,但Con M-ACF1:5作用相反。ConM-NCF和ConM-ACF都能抑制干細胞的心肌細胞分化能力,但又各有特點:ConM-NCF 1:50抑制早期心肌分化但不影響晚期心肌細胞標記物的表達;ConM-ACF 1:50抑制早期心肌分化比ConM-NCF 1:50更顯著,阻礙心肌成熟并上調分化晚期的心肌細胞標記物表達水平。此外,IL6中和抗體抑制ConM-...
