【摘要】：探討骨髓間充質(zhì)干細(xì)胞(BMSCs)移植在卵巢早衰小鼠卵巢功能重建中的應(yīng)用價(jià)值。通過一次性腹腔注射環(huán)磷酰胺和白消安建立小鼠卵巢早衰(POF)模型,并隨機(jī)分為建模組、BMSCs移植組和空白對(duì)照組。移植組于建模14 d經(jīng)尾靜脈注射綠色熒光蛋白(GFP)轉(zhuǎn)基因鼠BMSCs懸液(5×10~7個(gè)·mL~(-1)),每4 d重復(fù)1次,共5次;建模組和空白對(duì)照組注射等量生理鹽水。觀察建模后及BMSCs移植后小鼠的一般情況、血清FSH和E2水平、卵巢組織學(xué)變化以及GFP熒光信號(hào)在卵巢內(nèi)的分布。結(jié)果發(fā)現(xiàn),建模組小鼠食欲明顯減退,活動(dòng)減少,與空白對(duì)照組相比體重顯著減輕(P0.05),FSH水平顯著增高(P0.05),E2水平顯著降低(P0.05),卵巢體積減小,間質(zhì)纖維化嚴(yán)重,始基卵泡、生長卵泡和成熟卵泡數(shù)均顯著減少(P0.05)。BMSCs移植后,移植組小鼠體重增加,食欲增強(qiáng),活動(dòng)增多;28 d后FSH濃度顯著低于建模組(P0.05),E2濃度、始基卵泡、生長卵泡數(shù)及成熟卵泡數(shù)均顯著高于建模組(P0.05),但各項(xiàng)指標(biāo)仍與空白對(duì)照組間存在顯著性差異;熒光顯微鏡下可見綠色熒光信號(hào)散在分布于卵泡周圍。此結(jié)果提示,BMSCs可在小鼠損傷卵巢組織中定位存活,對(duì)卵巢組織結(jié)構(gòu)及內(nèi)分泌功能有修復(fù)作用。
[Abstract]:To investigate the application value of bone marrow mesenchymal stem cells (BMSCs) transplantation in ovarian function reconstruction of premature ovarian failure mice, the model of premature ovarian failure (POF) was established by intraperitoneal injection of cyclophosphamide (cyclophosphamide) and BMSCs. The BMSCs suspension of GFP transgenic mice was injected into caudal vein for 14 days, and it was repeated once every 4 days for 5 times. The normal saline was injected into the model group and the blank control group. The general situation, the serum FSH and E2 levels, the histological changes of ovary and the distribution of fluorescence signal of GFP in the ovary were observed after modeling and after BMSCs transplantation. In the model group, the appetite decreased and the activity decreased. Compared with the blank control group, the body weight of the model group decreased significantly. Compared with the control group, the level of P0.05FSH increased significantly. The level of E2 decreased significantly. The volume of ovaries decreased, the interstitial fibrosis was serious, and the primary follicles were also decreased. The number of growth follicles and mature follicles decreased significantly after transplantation, the body weight and appetite were increased in the transplanted group, and the FSH concentration was significantly lower than that in the model group at 28 days after transplantation, and the primary follicles were significantly lower than those in the model group. The number of growth follicles and mature follicles were significantly higher than that of the model group (P0.05A), but there were significant differences between the indexes of the model group and the blank control group. The green fluorescence signal was scattered around the follicles under the fluorescence microscope. The results suggested that BMSCs could survive in the damaged ovary tissue and repair the structure and endocrine function of the ovary.
