【摘要】：【目的】分析棉花黃萎病致病菌相關(guān)基因的功能揭示其中致病機(jī)制。【方法】通過ATMT法構(gòu)建了大麗輪枝菌(Verticillium dahliae)650個(gè)T-DNA插入突變體。利用hiTAIL-PCR技術(shù)獲得17株T-DNA插入體側(cè)翼基因序列,通過blast比對分析表明,其中16個(gè)基因序列編碼酶或蛋白。【結(jié)果】突變體的生物學(xué)性狀與野生型菌株Vd991相比,出現(xiàn)黃色菌絲型2株,中間菌絲型6株;部分突變體在生長速率(7株)、產(chǎn)孢量(14株)和粗毒素產(chǎn)量(4株)上都發(fā)生了具有統(tǒng)計(jì)學(xué)意義的變化(p0.05);致病力測定表明,9株突變體的致病力較野生型有所降低,在p0.05水平具有統(tǒng)計(jì)學(xué)意義。【結(jié)論】該研究為大麗輪枝菌致病基因的篩選和鑒定提供了理論基礎(chǔ)。
[Abstract]:[objective] to analyze the function of genes associated with Verticillium dahlia in cotton to reveal the pathogenic mechanism. [methods] 650 T-DNA insertion mutants of Verticillium dahlia were constructed by ATMT method, and 17 T-DNA inserts were obtained by hiTAIL-PCR technique. According to blast comparison analysis, 16 of them encode enzyme or protein. [results] compared with wild-type strain Vd991, two yellow mycelia and six intermediate mycelia were found in the mutant. The pathogenicity of 9 mutants was lower than that of wild type, and there were significant changes in growth rate (7 strains, sporulation: 14 strains) and crude toxin yield (4 strains) in some mutants (P < 0.05), the pathogenicity test showed that the pathogenicity of 9 mutants was lower than that of wild-type mutants, and the pathogenicity of 9 mutants was lower than that of wild-type mutants. There was statistical significance at p0.05 level. [conclusion] this study provides a theoretical basis for screening and identification of pathogenic genes of Trichoderma grandis.
