密閉微波輔助提取-HPLC法測(cè)定還亮草中硬飛燕草堿和巴比翠雀堿的含量

【摘要】:采用密閉微波輔助法(PMAE)提取還亮草中的硬飛燕草堿和巴比翠雀堿。采用單因素試驗(yàn)結(jié)合正交試驗(yàn)方法對(duì)微波實(shí)驗(yàn)條件進(jìn)行優(yōu)化。得到硬飛燕草堿和巴比翠雀堿的最佳提取方案:藥物顆粒度100目,固液比1∶60,微波溫度80℃,微波功率560 W,微波時(shí)間10 min。以甲醇-0.2%三乙胺(45∶55)為流動(dòng)相,建立了高效液相色譜(HPLC)測(cè)定硬飛燕草堿和巴比翠雀堿含量的方法。硬飛燕草堿和巴比翠雀堿分別在0.50~50.0,0.30~30.0 mg/L范圍內(nèi)呈良好的線性關(guān)系,回收率為98.3%~104.5%,相對(duì)標(biāo)準(zhǔn)偏差(RSD)分別為2.0%和2.2%。與傳統(tǒng)溶劑回流法(SRE)進(jìn)行比較,該方法簡(jiǎn)單、提取率更高。
