土壤中萘降解菌的耐鉛性能及機(jī)制初步研究

【摘要】:目的探討土壤中萘降解菌對萘和重金屬雙重污染的耐受性。方法對油田石油污染土壤中分離到的一株高效萘降解菌——伯克霍爾德菌(Burkholderia sp.)TN的重金屬耐受性、鉛富集機(jī)制及在土壤生物修復(fù)中的應(yīng)用進(jìn)行研究,以分光光度法測定不同Pb~(2+)濃度(100~500 mg/L)對菌懸液吸光度的影響,以電感耦合等離子體發(fā)射光譜儀(ICP-OES)測定培養(yǎng)基中重金屬(50 mg/L,Pb~(2+),Zn~(2+),Cu~(2+)和Cd~(3+))在菌體培養(yǎng)液、細(xì)胞壁、細(xì)胞內(nèi)富集物中的分布,以掃描電鏡(SEM)、能譜儀(EDS)和傅里葉變換紅外光譜(FTIR)觀察100 mg/L Pb~(2+)對菌體形態(tài)、元素及官能團(tuán)分布的影響。結(jié)果菌株TN對Pb~(2+)的最高耐受濃度為500 mg/L并可耐受一定濃度的Zn~(2+)、Cu~(2+)和Cd~(3+),TN對Pb~(2+)富集程度最高,細(xì)胞壁及胞內(nèi)富集率達(dá)到98.33%;菌體蓄積Pb~(2+)后細(xì)胞壁表面有沉淀物附著并有鉛元素檢出,對Pb~(2+)的蓄積主要依靠細(xì)胞壁上的羧基、羥基、酰胺基及磷酸基團(tuán);將TN菌株接種到含有2.5 mg/g萘及200 mg/L Pb~(2+)的滅菌土壤中,經(jīng)過13 d室溫培養(yǎng)之后,萘的去除率為94.29%。結(jié)論 TN菌株對Pb~(2+)、Zn~(2+)、Cu~(2+)和Cd~(3+)均具有一定的耐受性,對Pb~(2+)的富集能力強(qiáng)。該菌株能夠依靠細(xì)胞壁上的活性基團(tuán)富集Pb~(2+),適用于萘和Pb~(2+)雙重脅迫的土壤修復(fù)。
