【摘要】：目的探討重慶市在校大學(xué)生精液常規(guī)參數(shù)與精子DNA損傷水平之間的關(guān)聯(lián)性。方法 2014年6月招募的重慶大學(xué)生男性生殖健康調(diào)查(male reproductive health in Chongqing college students,MARHCS)隊(duì)列639名健康大學(xué)生,手淫法采集精液樣本,用計(jì)算機(jī)輔助精子分析系統(tǒng)(Computer-aided sperm analysis,CASA)進(jìn)行精液常規(guī)分析,采用Diff-Quick試劑盒檢測(cè)精子形態(tài),采用單細(xì)胞凝膠電泳(彗星實(shí)驗(yàn))檢測(cè)精子DNA損傷,采用Spearman相關(guān)分析探討精子DNA損傷與精液常規(guī)參數(shù)間的關(guān)聯(lián)性。結(jié)果大學(xué)生人群精液常規(guī)各指標(biāo)(中位數(shù)):精液量為3.6 m L,精子密度為52.0×106/m L,精子總活力為89.4%,前向運(yùn)動(dòng)精子百分率為57.0%,精子正常形態(tài)率為21.3%。精子DNA損傷(彗星實(shí)驗(yàn)指標(biāo))水平:尾部DNA含量百分比(Tail DNA%)為19.4%,尾長(zhǎng)(tail length,TL)為35.7 pixels,彗星長(zhǎng)(comet length,CL)為81.6 pixels,尾距(tail moment,TM)為7.6,Olive尾距(olive tail moment,OTM)為6.3。經(jīng)Spearman相關(guān)性分析,精子密度與Tail DNA%呈負(fù)向關(guān)聯(lián)(P0.05),精子總活力(%)與Tail DNA%、CL、TM、OTM呈負(fù)向關(guān)聯(lián)(P值均0.01),前向運(yùn)動(dòng)精子百分率與精子Tail DNA%、CL、TL、TM、OTM呈負(fù)向關(guān)聯(lián)(P值均0.05)。結(jié)論重慶市大學(xué)生精子密度、精子總活力、前向運(yùn)動(dòng)精子百分率均與精子DNA損傷呈負(fù)向關(guān)聯(lián)。
[Abstract]:Objective to investigate the relationship between sperm routine parameters and sperm DNA damage level in college students in Chongqing. Methods A total of 639 male reproductive health in Chongqing college students were enrolled in the male reproductive health survey in Chongqing in June 2014. Semen samples were collected by masturbation, and semen routine analysis was performed by computer-aided sperm analysis system (Computer-aided sperm Analysis CASAA). Diff-Quick kit was used to detect sperm morphology, single cell gel electrophoresis (comet assay) was used to detect sperm DNA damage, and Spearman correlation analysis was used to study the relationship between sperm DNA damage and semen parameters. Results the parameters of semen routine (median: sperm volume 3.6 mL, sperm density 52.0 脳 10 6 / mL, total sperm motility 89.4), forward motility sperm percentage 57.0%, normal sperm morphology rate 21.3% were found in the college students (median sperm volume 3.6 mL), sperm density (52.0 脳 10 6 / mL), total sperm motility (89 .4), forward motility sperm percentage (57.0%) and normal sperm morphology. Sperm DNA damage (comet assay index): the percentage of tail DNA content and tail length were 19.4, 35.7 pixels, 81.6 pixels and 7.6 OTM respectively. According to Spearman correlation analysis, there was a negative correlation between sperm density and Tail% (P0.05, total sperm motility) and Tail Tail Tail TMOTM (P = 0.01). The percentage of forward motile spermatozoa was negatively correlated with Tail TLT TLT ~ (TM) OTM (P = 0.05). Conclusion the sperm density, total sperm motility and the percentage of forward motility spermatozoa were negatively correlated with sperm DNA damage in Chongqing university students.
