復合重組磷脂酶用于大豆油脫膠的工藝優(yōu)化

【摘要】:利用實驗室前期構建的重組大腸桿菌所產磷脂酶A_1(phospholipase A_1,PLA_1)和磷脂酶C (phospholipase C,PLC)進行大豆油酶法脫膠研究,探討自主開發(fā)重組酶進行酶法脫膠的可行性。以諾維信商品酶Lecitase Ultra~(TM)為對照,研究酶法脫膠反應溫度、反應pH值、反應時間、攪拌速率、復合磷脂酶添加量工藝參數對大豆油脫膠的影響,并采用正交試驗對脫膠工藝條件進行優(yōu)化。研究結果表明,大豆油復合酶法脫膠的最佳工藝參數為:反應溫度45℃、反應pH 6.5、反應時間3 h、攪拌速率300 r/min、PLA_1和PLC添加量分別為7 940 U/kg和23 130 U/kg。復合磷脂酶對大豆油脫膠的效果與諾維信商品酶Lecitase Ultra~(TM)基本一致,大豆油磷含量可降至5 mg/kg以下,能夠滿足物理精煉的要求,為進一步開發(fā)具有知識產權的食品級油脂脫膠用酶制劑產品提供了理論依據。
[Abstract]:The enzymatic degumming of soybean oil was carried out by using phospholipase A1-PLA1 and C (phospholipase, and the feasibility of enzymatic degumming of recombinant Escherichia coli was discussed. The effects of enzymatic degumming reaction temperature, reaction pH value, reaction time, stirring rate and the amount of compound phospholipase on the degumming of soybean oil were studied.
