【摘要】：目的探討瑤藥小鉆乙酸乙酯提取物抗類風(fēng)濕關(guān)節(jié)炎(rheumatoid arthritis,RA)作用及相關(guān)機(jī)制。方法取雄性SD大鼠70只,隨機(jī)分為正常對(duì)照組、模型對(duì)照組、雷公藤組、地塞米松組、小鉆乙酸乙酯提取物低、中、高劑量組。建立膠原性關(guān)節(jié)炎(collagen induced arthritis,CIA)大鼠模型,連續(xù)灌胃給藥28 d,觀察大鼠狀態(tài)、體重、足腫脹度、關(guān)節(jié)炎指數(shù)、踝關(guān)節(jié)病變;采用ELISA法測(cè)定血清白細(xì)胞介素-1β(IL-1β)、白細(xì)胞介素-6(IL-6)及腫瘤壞死因子-α(TNF-α)含量;采用實(shí)時(shí)熒光定量PCR法檢測(cè)關(guān)節(jié)滑膜組織NF-κB p65 mRNA表達(dá)。結(jié)果小鉆乙酸乙酯提取物能顯著抑制CIA大鼠足腫脹,降低血清IL-1β、IL-6、TNF-α及滑膜組織NF-κB p65 mRNA水平,作用強(qiáng)于雷公藤,弱于地塞米松,與劑量相關(guān)。結(jié)論小鉆有一定抗RA作用,其機(jī)制可能與NF-κB p65轉(zhuǎn)錄有關(guān)。
[Abstract]:Objective to investigate the anti-rheumatoid arthritis (rheumatoid arthritis,RA) effect of ethyl acetate extract from Chinese herbal medicine. Methods 70 male SD rats were randomly divided into normal control group, model control group, Tripterygium wilfordii group, dexamethasone group and low, middle and high dose groups. The rat model of collagen-induced arthritis (collagen induced arthritis,CIA) was established. The rats were administrated intragastrically for 28 days. The state, body weight, foot swelling, arthritis index and ankle lesion were observed. The levels of serum interleukin-1 尾 (IL-1 尾), interleukin-6 (IL-6) and tumor necrosis factor- 偽 (TNF- 偽) were measured by ELISA assay, and the expression of NF- 魏 B p65 mRNA in synovial tissue was detected by real-time fluorescence quantitative PCR method. Results the ethyl acetate extract significantly inhibited foot swelling in CIA rats, and decreased the levels of serum IL-1 尾, IL-6,TNF- 偽 and NF- 魏 B p65 mRNA in synovial tissue, which was stronger than Tripterygium wilfordii and weaker than that of dexamethasone in dose-dependent manner. Conclusion small drill has some anti-RA effect, and its mechanism may be related to the transcription of NF- 魏 B p65.
