MiR-193b介導(dǎo)的電針減少Stat5NKO肥胖小鼠體重的機(jī)制研究

【摘要】:目的:觀察電針對(duì)Stat5NK0小鼠的減肥效應(yīng)及與miR-193b的相關(guān)性。方法:選擇周齡在3-4周的Stat5fl/fi 和 Statffl/fl;Nestin-cre (StatfNKO)雄性小鼠,所有小鼠均喂食普通標(biāo)準(zhǔn)飼料,記錄體重,以體重大于等于Stat5fl/fl組小鼠平均體重20%的標(biāo)準(zhǔn)入選SSttat5NKO組。再把Stat5ffl/fl和Stat5NKO小鼠隨機(jī)分為對(duì)照組和針刺組(Stat5fl/fl+EA;Stat5NO+EA)。使用特殊工具將小鼠固定,并在小鼠清醒狀態(tài)下進(jìn)行電針干預(yù),電針穴取小鼠“后三里”和“內(nèi)庭”,對(duì)照組只捆不針。每周記錄小鼠體重。在電針治療前后,分別對(duì)小鼠進(jìn)行糖耐量實(shí)驗(yàn)(IGTT)。電針治療結(jié)束后,進(jìn)行冷庫(kù)實(shí)驗(yàn)。處死小鼠,取血清檢測(cè)瘦素、胰島素、甘油三酯、膽固醇和低密度脂蛋白;取小鼠附睪脂肪并稱重。使用蘇木精一伊紅染色法(hematoxylin-eosinstaining、HE Staining)研究針刺對(duì)脂肪組織的作用。使用實(shí)時(shí)熒光定量核酸擴(kuò)增檢測(cè)系統(tǒng)(real-time quantitative PCR detecting system, qPCR)檢測(cè)miR-193b在小鼠不同組織中的表達(dá)差異,檢測(cè)解耦聯(lián)蛋白(UCP1)在骨骼肌與附睪脂肪中的表達(dá)水平。結(jié)果:電針有效降低了肥胖小鼠的體重,體脂比顯著低于對(duì)照組,電針組小鼠糖耐量有一定程度改善,血脂生化指標(biāo)顯著改善,耐寒性得到提高。HE染色可見電針組小鼠附睪脂肪細(xì)胞體積減小。qPCR實(shí)驗(yàn)證實(shí),miR-193b在小鼠的棕色脂肪中含量最為豐富;Stat5NKO+EA組小鼠外周組織中miR-193b的表達(dá)下降,而在中樞表達(dá)上升;電針能促進(jìn)UCP1在骨骼肌和附睪脂肪中的表達(dá)。結(jié)論:電針能降低5Stat5NKO肥胖小鼠的體重;miR-193b在小鼠棕色脂肪中表達(dá)最為豐富;電針能調(diào)控miR-193b在組織中的表達(dá),這可能需要中樞Stat5的參與。
[Abstract]:Objective: to observe the effect of electroacupuncture on the weight loss of Stat5NK0 mice and its correlation with miR-193b. Methods: Stat5fl-fi and Statflt-nestin-cre (StatfNKO) male mice aged 3-4 weeks were selected. All the mice were fed with normal diet and their body weight was recorded. The average body weight of SSttat5NKO group was 20% or more than that of Stat5fl-fl group. Stat5ffl / fl and Stat5NKO mice were randomly divided into control group and acupuncture group. The mice were fixed with special tools, and electroacupuncture intervention was carried out in the awake state of the mice. The mice were taken from the electroacupuncture points for "three li" and "chambers", while the control group was not tied with the needles. The weight of mice was recorded weekly. Glucose tolerance test (IGTT) was performed on mice before and after electroacupuncture treatment. After electroacupuncture treatment, cold storage experiment was carried out. Serum leptin, insulin, triglyceride, cholesterol and low density lipoprotein were measured, and epididymal fat was taken and weighed. The effect of acupuncture on adipose tissue was studied by hematoxylin-eosin stenting (HE Staining). Real-time quantitative detecting system (qPCR) was used to detect the expression of miR-193b in different tissues of mice and the expression level of UCP1 in skeletal muscle and epididymal fat. Results: electroacupuncture effectively reduced the body weight of obese mice, the body fat ratio was significantly lower than that of the control group, the glucose tolerance of the mice in the electroacupuncture group was improved to a certain extent, and the biochemical indexes of blood lipids were significantly improved. The enhanced cold tolerance. He staining showed that the volume of epididymal adipocytes in electroacupuncture group was reduced. QPCR results showed that miR-193b was the most abundant in brown fat of mice. The expression of miR-193b in peripheral tissues of mice in Stat5NKO EA group decreased, but increased in the center of mice. Electroacupuncture can promote the expression of UCP1 in skeletal muscle and epididymal fat. Conclusion: electroacupuncture can reduce the expression of miR-19b in brown fat of 5Stat5NKO obese mice, and electroacupuncture can regulate the expression of miR-193b in tissues, which may require the participation of central Stat5.
