【摘要】：目的觀察經(jīng)后增殖方對超排卵小鼠卵巢凋亡相關(guān)基因及其蛋白表達的調(diào)節(jié)作用,探討中藥益氣血法對超排卵小鼠卵巢顆粒細胞凋亡及卵泡質(zhì)量的影響。方法建立超排卵小鼠模型,30只小鼠隨機分為空白組、模型組、中藥治療組。采用實時熒光定量PCR(real-time quantitative PCR,qPCR)和免疫印跡法(Western blot)分別檢測Bcl-2、Bax、Caspase3基因和蛋白的表達水平。結(jié)果中藥治療組Bax、Cas-pase3 mRNA的表達水平與模型組比較(0.895 vs 1.779,0.674 vs 2.066,均P0.05),差異均有統(tǒng)計學(xué)意義;中藥治療組Bcl-2、Bax、Caspase3 mRNA的表達水平與空白組比較(1.11 vs 1.26,0.89 vs 0.79,0.67 vs 0.96),差異均無統(tǒng)計學(xué)意義(P0.05);中藥治療組Bcl-2、Bax、Caspase3蛋白的表達水平與模型組比較(0.62 vs0.41,0.79 vs 1.06,0.49 vs 0.89),差異均有統(tǒng)計學(xué)意義(P0.05),與空白組比較(0.62 vs 0.64,0.79 vs 0.73,0.49 vs 0.50)差異均無統(tǒng)計學(xué)意義(P0.05)。結(jié)論經(jīng)后增殖方能促進Bcl-2蛋白的表達,抑制Bax、Cas-pase3基因及蛋白的表達,抑制超排卵小鼠卵巢顆粒細胞凋亡至接近自然排卵小鼠的水平,提高超排卵小鼠卵泡質(zhì)量。
[Abstract]:Objective to observe the effect of Jinghou recipe on the expression of ovarian apoptosis-related genes and proteins in superovulation mice, and to explore the effect of Yiqi and Xue method on ovarian granulosa cell apoptosis and follicle quality in superovulation mice. Methods Thirty mice with hyperovulation were randomly divided into blank group, model group and traditional Chinese medicine treatment group. The expression of caspase3 gene and protein were detected by real-time quantitative qPCR and Western blotting, respectively. Results compared with the model group, the expression level of Bax-Caspase3 mRNA in the traditional Chinese medicine treatment group was 0.895 vs 1.779c0.674 vs 2.066, all the differences were statistically significant (P 0.05). The expression level of Bcl-2OBax-Caspase3 mRNA in the TCM treatment group was higher than that in the control group (1.11 vs 1.260.89 vs 0.79U 0.67 vs 0.96), and there was no significant difference in the expression of Bcl-2Bax-Caspase3 protein between the Chinese medicine group and the model group (0.62 vs 0.41 vs 1.06 鹵0.49 vs 0.89, respectively), and the difference was statistically significant (P 0.05P 0.05A). The expression of Bcl-2AXCaspase3 mRNA in the treatment group was significantly higher than that in the control group (P 0.05). Compared with the control group, there was no significant difference in 0.62 vs 0.64N 0.79 vs 0.73N 0.49 vs 0.50). Conclusion the prescription can promote the expression of Bcl-2 protein, inhibit the expression of Bax-Cas-pase3 gene and protein, inhibit the apoptosis of ovarian granulosa cells to the level of natural ovulation mice, and improve the follicle quality of superovulation mice.
