【摘要】：目的:分析脾虛肝癌小鼠有機(jī)陰離子轉(zhuǎn)運(yùn)肽2a1(Oatp2a1)m RNA和蛋白的表達(dá)情況,探討脾虛因素對(duì)肝癌的影響及其機(jī)制。方法:60只小鼠分成正常對(duì)照組、原位移植肝癌組和脾虛原位移植肝癌組,建立脾虛原位移植肝癌小鼠模型,測(cè)量各組肝癌腫瘤的體積,采用RT-PCR和Western Blot的方法檢測(cè)小鼠各組織中Oatp2a1的表達(dá)。結(jié)果:脾虛原位移植肝癌組小鼠肝癌腫瘤體積明顯大于原位移植肝癌組(P0.01),肝癌造模后小鼠各組織Oatp2a1的表達(dá)發(fā)生了變化,癌組織中Oatp2a1表達(dá)均明顯高于相應(yīng)組肝組織(P0.01),與原位移植肝癌組小鼠比較,脾虛原位移植肝癌組小鼠胃、肝和脾組織Oatp2a1 m RNA表達(dá)明顯降低(P0.01),癌組織表達(dá)則明顯升高(P0.01)。結(jié)論:脾虛因素能促進(jìn)小鼠肝癌的發(fā)生和發(fā)展,其機(jī)制可能是脾虛內(nèi)環(huán)境下各組織Oatp2a1的異常表達(dá),進(jìn)一步提示脾虛濕濁轉(zhuǎn)運(yùn)障礙與肝癌密切相關(guān)。
[Abstract]:Objective: to investigate the effect of spleen deficiency on liver cancer and its mechanism by analyzing the expression of mRNA and protein of organic anion transporter peptide 2a1Oatp2a1in mice with liver cancer due to spleen deficiency. Methods 60 mice were divided into normal control group, orthotopic liver transplantation group and spleen deficiency orthotopic transplantation group. The expression of Oatp2a1 in mouse tissues was detected by RT-PCR and Western blot. Results: the tumor volume of liver cancer in spleen deficiency orthotopic liver transplantation group was significantly larger than that in orthotopic transplantation liver cancer group (P 0.01). The expression of Oatp2a1 in various tissues of mice was changed after liver cancer model was established. The expression of Oatp2a1 in liver tissue was significantly higher than that in liver tissue of corresponding group. Compared with orthotopic liver transplantation group, the expression of Oatp2a1 mRNA in stomach, liver and spleen tissue of liver cancer group with spleen deficiency was significantly lower than that in liver tissue of liver cancer group, while the expression of Oatp2a1 mRNA in cancer tissue was significantly higher than that in orthotopic liver transplantation group. Conclusion: spleen deficiency can promote the occurrence and development of liver cancer in mice, and its mechanism may be the abnormal expression of Oatp2a1 in various tissues under the internal environment of spleen deficiency, which further indicates that the obstacle of transferring dampness due to spleen deficiency is closely related to liver cancer.
