【摘要】：實(shí)驗(yàn)?zāi)康?基于“共同土壤”學(xué)說(shuō)探索建立2型糖尿病合并冠心病大鼠動(dòng)物模型,并根據(jù)“寒獨(dú)留則血凝泣,凝則脈不通”的中醫(yī)理論在前者基礎(chǔ)上進(jìn)一步建立2型糖尿病合并冠心病寒凝血瘀證大鼠模型。實(shí)驗(yàn)方法:運(yùn)用高脂高糖飼料喂養(yǎng)小劑量腹腔注射鏈脲佐菌素(STZ)以及維生素D3腹腔注射并加上冰浴方法進(jìn)行造模。應(yīng)用量化評(píng)分表對(duì)造模期間大鼠的癥狀和體征進(jìn)行評(píng)分,并在造模的第30、60、90天以及注射STZ后行眼眶采血,用全自動(dòng)生化儀以及血脂檢測(cè)試劑盒測(cè)定大鼠血脂,用血糖檢測(cè)儀檢測(cè)大鼠空腹血糖,用全自動(dòng)血流變檢測(cè)血液流變學(xué)改變以及用ELESA法檢測(cè)大鼠血清胰島素、前列腺素和血栓素的變化,用心電圖機(jī)記錄大鼠心電圖Ⅱ?qū)?lián)ST段的改變,并于造模結(jié)束后對(duì)大鼠的心臟與胰腺進(jìn)行HE染色以觀察其組織形態(tài)的改變。實(shí)驗(yàn)結(jié)果:(1)模型組大鼠在癥狀與體征方面出現(xiàn)毛發(fā)豎立、枯萎,畏寒喜暖,喜愛扎堆,行動(dòng)遲鈍,活動(dòng)減少,飲水以及攝食量均減少,體重減輕,大便稀軟,耳緣、爪甲紫暗,舌質(zhì)暗紅有瘀斑,舌下絡(luò)脈曲張等寒凝血瘀證癥狀。(2)模型組大鼠血脂、血糖、全血黏度均增高,與空白組對(duì)照有顯著性差異(P0.05,P0.01)。(3)模型組大鼠心電圖II導(dǎo)聯(lián)ST段有顯著下移現(xiàn)象,與空白組對(duì)照有顯著性差異(P0.01)(4)模型組大鼠血清胰島素、前列腺素、血栓素均出現(xiàn)紊亂,與空白組對(duì)照有顯著性差異(P0.05,P0.01)。(5)模型組大鼠心臟組織出現(xiàn)冠狀動(dòng)脈硬化、心肌細(xì)胞水腫、壞死,胰腺組織出現(xiàn)胰島細(xì)胞萎縮破壞等病理學(xué)改變。實(shí)驗(yàn)結(jié)論:運(yùn)用高脂高糖飼料喂養(yǎng)小劑量腹腔注射鏈脲佐菌素(STZ)以及維生素D3灌胃并加上冰浴方法建立的Ⅱ型糖尿病合并冠心病寒凝血瘀證與中醫(yī)證侯特點(diǎn)相符合,具有一定的理論和實(shí)踐意義。
[Abstract]:Objective: to establish an animal model of type 2 diabetes mellitus with coronary heart disease (CHD) based on the theory of "common soil". On the basis of the former, the model of type 2 diabetes mellitus with cold coagulation and blood stasis syndrome of coronary heart disease was established. Methods: low dose of STZ and vitamin D3 were fed with high fat and high sugar diet, and the model was made by ice bath. The symptoms and signs of the rats during model making were scored by using the quantitative scoring table. The blood samples were collected from the orbit on the 30th day of model making and after STZ injection. The blood lipid of the rats was measured by automatic biochemical instrument and blood lipid test kit. Fasting blood glucose was detected by blood glucose detector, hemorheology was detected by automatic hemorheology and serum insulin, prostaglandin and thromboxane were detected by ELESA method.
