【摘要】：目的:通過觀察慢性再生障礙性貧血(Chronic aplastic anemia,CAA,以下簡稱"慢性再障”,中醫(yī)稱"慢性髓勞病")患者服用補(bǔ)髓生血顆粒前后骨髓單個(gè)核細(xì)胞中整合素連接激酶(Integrin-linked kinase,ILK)蛋白表達(dá)的變化,探討補(bǔ)髓生血顆粒對(duì)再障患者的ILK及造血微環(huán)境的影響,進(jìn)而了解ILK與再障發(fā)生發(fā)展的關(guān)系,為探索補(bǔ)髓生血顆粒治療慢性再障的機(jī)理提供依據(jù)。方法:1.收集30例應(yīng)用補(bǔ)髓生血顆粒治療的慢性再障(CAA)患者的骨髓;2.收集10例健康人的骨髓;3.采用Western blot法檢測(cè)患者治療前后以及與健康對(duì)照組骨髓單個(gè)核細(xì)胞ILK蛋白的表達(dá)水平。結(jié)果:1.補(bǔ)髓生血顆粒治療慢性髓勞病的臨床療效達(dá)到80.00%,療效較確切。2.慢性髓勞病患者治療前ILK蛋白表達(dá)水平明顯高于健康組,差異有統(tǒng)計(jì)學(xué)意義(P0.05),治療組經(jīng)補(bǔ)髓生血顆粒治療后,ILK蛋白表達(dá)水平較治療前顯著下降,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。3.治療組治療后的ILK蛋白表達(dá)水平仍然是高于健康對(duì)照組的,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:1.補(bǔ)髓生血顆粒對(duì)慢性髓勞病有一定治療作用是可以肯定的。2.慢性髓勞患者ILK蛋白高于正常人表達(dá)水平可能參與慢性髓勞的發(fā)病過程。3.補(bǔ)髓生血顆�？赡芡ㄟ^降低慢性髓勞患者ILK蛋白的表達(dá)水平以調(diào)節(jié)整合素β1介導(dǎo)的造血微環(huán)境損傷,進(jìn)而促進(jìn)造血干/祖細(xì)胞的增殖分化以達(dá)到治療慢性髓勞病的目的。
[Abstract]:Objective: to observe the expression of integrin linked kinase integrin-linked kinase (ILK) protein in bone marrow mononuclear cells of chronic aplastic anemia patients with chronic aplastic anemia ("chronic aplastic anemia") before and after taking "chronic myelopathy".To explore the effect of Bupuishengxue granule on ILK and hematopoietic microenvironment in patients with aplastic anemia, and to understand the relationship between ILK and the occurrence and development of aplastic anemia, and to provide the basis for exploring the mechanism of Busuishengxue granule in treating chronic aplastic anemia.Method 1: 1.The bone marrow of 30 patients with chronic aplastic anemia (CAA) treated with Bupu Shengxue granule was collected.The bone marrow of 10 healthy persons was collected.The expression of ILK protein in bone marrow mononuclear cells before and after treatment and control group was detected by Western blot assay.The result is 1: 1.The clinical curative effect of Busui Shengxue granule in treating chronic myellabour disease is 80.00g, and the curative effect is more accurate. 2. 2.The expression level of ILK protein was significantly higher in patients with chronic myellabour disease before treatment than that in healthy control group (P 0.05), and the expression level of ilk protein in treatment group was significantly lower than that before treatment (P 0.05 路3).The expression of ILK protein in the treatment group was still higher than that in the healthy control group (P 0.05).Conclusion 1.Busui Shengxue granule has certain therapeutic effect on chronic myellabour disease. 2. 2.The expression level of ILK protein in patients with chronic myelosis may be involved in the pathogenesis of chronic myelosis. 3. 3.Busui Shengxue granule may regulate integrin 尾 1-mediated hematopoietic microenvironment injury by decreasing the expression level of ILK protein in chronic pull-stress patients, and then promote the proliferation and differentiation of hematopoietic stem / progenitor cells to achieve the purpose of treating chronic myelo disease.
