電針聯(lián)合跑臺訓(xùn)練對大鼠骨骼肌鈍挫傷的修復(fù)作用

【摘要】:目的:觀察電針聯(lián)合跑臺訓(xùn)練對大鼠骨骼肌鈍挫傷的修復(fù)作用,探討電針聯(lián)合跑臺訓(xùn)練改善骨骼肌損傷的可能機(jī)制。方法:SD大鼠隨機(jī)分為正常組、模型組、電針組、跑臺組和聯(lián)合組,每組12只。采用自制打擊裝置建立骨骼肌鈍挫傷模型。電針組電針"足三里"、阿是穴,跑臺組進(jìn)行跑臺訓(xùn)練,聯(lián)合組同時(shí)進(jìn)行電針和跑臺訓(xùn)練,1次/d,連續(xù)至損傷后3、14d。HE染色觀察損傷后14d腓腸肌新生肌細(xì)胞橫截面積和直徑,Western blot法檢測損傷后3d腓腸肌哺乳動(dòng)物雷帕霉素靶蛋白(mTOR)、肌細(xì)胞生成素(myoG)和14d腓腸肌mTOR、快肌型骨骼肌肌球蛋白重鏈(Fast MyHC)的表達(dá)量。結(jié)果:模型組新生肌細(xì)胞橫截面積和直徑明顯小于正常組(P0.01),各治療組均顯著大于模型組(P0.05),聯(lián)合組顯著大于電針組和跑臺組(P0.05)。與正常組比較,損傷后3d模型組腓腸肌mTOR和myoG蛋白表達(dá)均顯著升高(P0.01);與模型組比較,各治療組腓腸肌mTOR和myoG蛋白表達(dá)均顯著升高(P0.01);與電針組比較,跑臺組腓腸肌mTOR和myoG蛋白表達(dá)均顯著降低(P0.01);與電針組和跑臺組比較,聯(lián)合組腓腸肌mTOR和myoG蛋白表達(dá)顯著升高(P0.05,P0.01)。與正常組比較,損傷后14d模型組腓腸肌mTOR蛋白表達(dá)升高,但差異無統(tǒng)計(jì)學(xué)意義(P0.05),Fast MyHC蛋白表達(dá)顯著降低(P0.01);與模型組比較,各治療組腓腸肌mTOR及Fast MyHC蛋白表達(dá)均顯著升高(P0.05,P0.01);與電針組比較,跑臺組腓腸肌mTOR及Fast MyHC蛋白表達(dá)均顯著降低(P0.05,P0.01);與電針組和跑臺組比較,聯(lián)合組腓腸肌mTOR及Fast MyHC蛋白表達(dá)均顯著升高(P0.01)。結(jié)論:電針聯(lián)合跑臺訓(xùn)練能促進(jìn)成肌細(xì)胞分化成熟,減輕骨骼肌損傷,該作用與增加mTOR表達(dá)量,正向調(diào)控myoG、Fast MyHC的表達(dá)有關(guān)。
[Abstract]:Objective: To observe the effect of electroacupuncture combined with rehabilitation training on the run and skeletal muscle contusion of rats, and explore the possible mechanism of electroacupuncture combined with treadmill training improves skeletal muscle injury. Methods: SD rats were randomly divided into normal group, model group, electroacupuncture group, running group and combination group, 12 rats in each group. Using the self-made striking device skeletal muscle contusion model. The electroacupuncture group and Electroacupuncture of "Zusanli", Ashi, treadmill treadmill training group, combination group and electroacupuncture and treadmill training, 1 /d, continuous to staining 14d after injury of gastrocnemius muscle cells of newborn cross-sectional area and the diameter of 3,14d.HE after injury, Western blot detection of 3D after injury of gastrocnemius muscle of the mammalian target of rapamycin (mTOR), myogenin (myoG) and 14d gastrocnemius mTOR, fast twitch skeletal muscle type myosin heavy chain (Fast MyHC) expression. Results: the model group of new muscle cell cross-sectional area and diameter significantly Less than the normal group (P0.01), the treatment group were significantly higher than the model group (P0.05), combination group was significantly higher than that in the electroacupuncture group and treadmill group (P0.05). Compared with the normal group, the expression of 3D after injury in model group gastrocnemius mTOR and myoG protein were significantly increased (P0.01); compared with the model group, the expression of the treatment group gastrocnemius mTOR and myoG protein were significantly increased (P0.01); compared with the electroacupuncture group, running group gastrocnemius mTOR and myoG protein expression were significantly decreased (P0.01); and the electroacupuncture group and treadmill group compared with gastrocnemius mTOR and myoG protein expression was significantly increased (P0.05, P0.01). With the normal group, model group, 14d expression of gastrocnemius muscle mTOR protein increased after injury, but the difference was not statistically significant (P0.05), the expression of Fast MyHC protein was significantly decreased (P0.01); compared with the model group, the expression of each treatment group mTOR and Fast MyHC protein in the gastrocnemius muscle were significantly increased (P0.05, P0.01); compared with the electroacupuncture group run, Taiwan Group mTOR and Fast gastrocnemius muscle MyHC protein expression were significantly decreased (P0.05, P0.01); and the electroacupuncture group and treadmill group compared with group mTOR and Fast gastrocnemius muscle MyHC protein expression increased significantly (P0.01). Conclusion: electroacupuncture combined with treadmill training can promote myoblast differentiation and maturation, reduce the injury of skeletal muscle this effect, with the increase of mTOR expression, the positive regulation of myoG, the expression of Fast MyHC.
