【摘要】：目的應(yīng)用基于色譜質(zhì)譜聯(lián)用技術(shù)(LC-MS)的蛋白質(zhì)組學(xué)鑒定篩選胃癌相關(guān)蛋白質(zhì),并初步探討其功能。方法選取2011年4月—2012年9月于河北醫(yī)科大學(xué)第四醫(yī)院確診為胃腺癌并進(jìn)行手術(shù)切除者16例為研究對(duì)象。術(shù)中取患者胃癌組織及癌旁正常組織,采用基于LC-MS的蛋白質(zhì)組學(xué)鑒定篩選胃癌組織及癌旁正常組織中的差異蛋白質(zhì),并應(yīng)用免疫組化染色法驗(yàn)證篩選出的差異蛋白質(zhì)。結(jié)果基于LC-MS的蛋白質(zhì)組學(xué)鑒定結(jié)果顯示,胃癌組織與癌旁正常組織均鑒定了3 000~4 000種蛋白質(zhì),其中12種蛋白質(zhì)差異顯著,將其與GO數(shù)據(jù)庫(kù)進(jìn)行對(duì)比,去除二級(jí)節(jié)點(diǎn)中歸類為"細(xì)胞組分"的結(jié)構(gòu)蛋白質(zhì),在"分子功能"和"代謝過(guò)程"這兩個(gè)節(jié)點(diǎn)中找到了4種可能與胃癌有關(guān)的蛋白質(zhì),即谷胱甘肽巰基轉(zhuǎn)移酶-π-1(GST-π-1)、銅鋅超氧化物歧化酶(Cu/Zn-SOD)、丙酮酸激酶M2(PK-M2)、P120連環(huán)蛋白(P120-ctn)。免疫組化染色結(jié)果顯示,胃癌組織中GST-π-1、PK-M2、P120-ctn陽(yáng)性率大于癌旁正常組織,Cu/Zn-SOD陽(yáng)性率小于癌旁正常組織(P0.001),與基于LC-MS的蛋白質(zhì)組學(xué)鑒定結(jié)果相符。結(jié)論基于LC-MS的蛋白質(zhì)組學(xué)鑒定可篩選出胃癌相關(guān)蛋白質(zhì)(GST-π-1、Cu/Zn-SOD、PK-M2、P120-ctn),初步分析認(rèn)為其與胃癌的多種細(xì)胞生物學(xué)活動(dòng)有關(guān),證實(shí)基于LC-MS的蛋白質(zhì)組學(xué)鑒定在胃癌相關(guān)蛋白質(zhì)篩選方面具有明顯的應(yīng)用價(jià)值。
[Abstract]:Objective to screen gastric cancer related proteins by proteomics based on LC-MS and to explore their functions. Methods from April 2011 to September 2012, 16 cases of gastric adenocarcinoma were diagnosed and surgically resected from the fourth Hospital of Hebei Medical University. The differentially expressed proteins in gastric cancer tissues and adjacent normal tissues were screened by proteomics identification based on LC-MS, and the differentially identified proteins were verified by immunohistochemical staining. Results the results of proteomics analysis based on LC-MS showed that 3 000 ~ 4 000 proteins were identified in gastric cancer tissues and adjacent normal tissues, among which 12 proteins had significant differences. The results were compared with GO database. Removing structural proteins classified as "cellular components" in secondary nodes, four proteins that might be associated with gastric cancer were found in the "molecular function" and "metabolic processes". They are glutathione mercaptotransferase-蟺 -1 (GST- 蟺 -1), copper zinc superoxide dismutase (Cu/Zn-SOD), pyruvate kinase M2 (PK-M2) and P120 catenin (P120-ctn). The results of immunohistochemical staining showed that the positive rate of GST- 蟺 -1 PK-M2P P120-ctn was higher than that of adjacent normal tissues, and the positive rate of Cu/Zn-SOD was lower than that of adjacent normal tissues (P0.001). The results were consistent with the results of proteomics identification based on LC-MS. Conclusion the proteome identification based on LC-MS can screen out GST- 蟺 -1CU / Zn-SODN-PK-M2P120-ctn, which is related to many cell biological activities of gastric cancer. It is confirmed that proteomics based on LC-MS is of great value in the screening of gastric cancer related proteins.
