EGCG對(duì)胃癌細(xì)胞SHH信號(hào)通路Ptch、Gli-1的表達(dá)影響

【摘要】:目的:探討(-)-Epigallocatechin gallate(EGCG)對(duì)胃癌細(xì)胞(AGS)增殖、凋亡影響,以及對(duì)Sonic hedgehog(SHH)信號(hào)通路相關(guān)蛋白Ptch、Gli-1的表達(dá)影響。方法:將AGS細(xì)胞株在37℃,5%CO2飽和濕度溫箱里進(jìn)行培養(yǎng)、換液和傳代。實(shí)驗(yàn)組EGCG濃度為25μg/ml、50μg/ml、100μg/ml,并設(shè)置陰性對(duì)照組,實(shí)驗(yàn)時(shí)取對(duì)數(shù)期細(xì)胞。應(yīng)用MTT法和流式細(xì)胞儀檢測(cè)EGCG對(duì)AGS胃癌細(xì)胞增殖與凋亡,細(xì)胞爬片免疫組化檢測(cè)SHH信號(hào)通路相關(guān)蛋白Ptch、Gli-1的表達(dá)影響。結(jié)果:EGCG干預(yù)AGS細(xì)胞48h后,隨著EGCG濃度升高,細(xì)胞貼壁數(shù)量減少且形態(tài)變圓,漂浮數(shù)量增多,分布變稀疏,可見(jiàn)到細(xì)胞崩解后的碎片、顆粒等;而陰性對(duì)照組AGS細(xì)胞貼壁良好,輪廓清晰。隨著EGCG濃度增高,AGE胃癌細(xì)胞增殖抑制率、凋亡率明顯提高(P0.05);隨著EGCG濃度的升高,Gli-1表達(dá)顯著降低而Ptch表達(dá)顯著升高(P0.05)。結(jié)論:EGCG可以抑制SHH信號(hào)通路的相關(guān)蛋白促使AGS胃癌細(xì)胞凋亡
