體外不完全射頻消融后肝癌細胞上皮-間質轉化相關長鏈非編碼RNA差異表達

【摘要】:目的研究上皮-間質轉化(EMT)相關長鏈非編碼RNA(LncRNA)在不完全射頻消融(RFA)肝細胞癌(HCC)病灶的差異表達譜變化。方法采用Huh7細胞47℃水浴加熱,獲取體外模擬的不完全RFA HCC細胞。鏡下及免疫印跡檢測Huh7-H細胞EMT改變,Transwell檢測細胞遷移與侵襲能力,CCK-8檢測細胞增殖能力。采用Lnc Path~(TM) Human EMT Array芯片分析Huh7-H和Huh7差異表達的EMT相關LncRNA,RT-PCR驗證。結果鏡下顯示Huh7-H呈現EMT形態(tài)學改變。免疫印跡檢測顯示Huh7-H細胞上皮表型標志分子E-cadherin表達降低,間質表型標志分子N-cadherin、vimentin表達增加。Transwell遷移及侵襲實驗顯示,Huh7-H、Huh7穿膜細胞數分別為(138.0±11.8)和(61.0±5.2)、(82.7±39.4)和(33.3±7.8),Huh7-H細胞遷移及侵襲能力明顯增強(P0.05)。CCK-8檢測顯示Huh7-H細胞增殖能力明顯增強(P0.05)。Lnc Path~(TM) Human EMT Array芯片篩選出3個差異表達LncRNA(P≤0.05,變化倍數≥1.5),即2條LncRNA(FUNDC2P4、RPL27P7)下調,1條LncRNA(MTND4LP14)上調。RT-PCR驗證HUH7-H組FUNDC2P4、RPL27P7、MTND4LP14基因表達豐度分別是HUH-7組的0.137、0.869、1.037倍。臨床標本驗證LncRNA FUNDC2P4在癌旁組織表達高于HCC組織,HCC組織高于RFA術后殘余HCC組織。結論篩選出不完全RFA后HCC細胞低表達的EMT相關LncRNA FUNDC2P4,為深入探討該基因功能及分子機制提供了基礎。
