【摘要】：目的觀察芒柄花黃素對(duì)人胃癌MKN-45細(xì)胞株增殖、凋亡的影響,并探討其可能作用機(jī)制。方法取對(duì)數(shù)生長(zhǎng)期人胃癌MKN-45細(xì)胞分為A、B、C、對(duì)照組,A、B、C組分別加入20、40、80μg/m L芒柄花黃素培養(yǎng),對(duì)照組加入不含芒柄花黃素的完全培養(yǎng)基。采用MTT法觀察給藥24、48、72 h各組細(xì)胞增殖情況,計(jì)算細(xì)胞增殖抑制率。采用DAPI染色法評(píng)價(jià)其對(duì)MKN-45細(xì)胞形態(tài)學(xué)的影響。采用Western blotting法檢測(cè)給藥48 h時(shí)各組細(xì)胞磷酸化IκB(p-IκB)及NF-κB p65。結(jié)果隨芒柄花黃素濃度升高,細(xì)胞增殖抑制率升高,且隨干預(yù)時(shí)間增加,細(xì)胞增殖抑制率升高(P均0.05)。給藥72 h時(shí)A、B、C、對(duì)照組的凋亡率分別為25.62%±2.57%、48.27%±3.18%、72.51%±4.35%、1.07%±0.54%,A、B、C組與對(duì)照組相比,P均0.05。給藥48 h時(shí)A、B、C組細(xì)胞p-IκB、NF-κB p65蛋白相對(duì)表達(dá)量均高于對(duì)照組,且隨芒柄花黃素濃度升高,細(xì)胞p-IκB及NF-κB p65蛋白相對(duì)表達(dá)量增加,P均0.05。結(jié)論芒柄花黃素可抑制人胃癌MKN-45細(xì)胞株的增殖、促進(jìn)細(xì)胞凋亡,其機(jī)制可能為芒柄花黃素激活NF-κB信號(hào)通路。
[Abstract]:Objective to observe the effect of Flavatin on proliferation and apoptosis of human gastric cancer MKN-45 cell line and to explore its possible mechanism. Methods Human gastric cancer MKN-45 cells at logarithmic growth stage were divided into two groups: control group and control group. The control group was cultured with 20 ~ 4080 渭 g / m ~ (-1) amaranthin, and the control group was cultured on a complete medium without Acanthoxanthin. MTT assay was used to observe the proliferation of the cells in each group for 72 h. The effect of DAPI staining on the morphology of MKN-45 cells was evaluated. Phosphorylated I 魏 B (p-I 魏 B) and NF- 魏 B p65 were detected by Western blotting assay at 48 h after administration. Results the inhibition rate of cell proliferation was increased with the increase of the concentration of anthocyanin, and the inhibition rate of cell proliferation was increased with the increase of intervention time (P 0.05). At 72 h after administration, the apoptotic rates in the control group were 25.62% 鹵2.57% 鹵48.27% 鹵3.18%, respectively, and 72.551% 鹵4.3551% 鹵0.54% in the control group, respectively, compared with those in the control group (P 0.05). The relative expression of p-I 魏 B and NF- 魏 B p65 protein in the cells of Agni BU C group was higher than that in the control group at 48 h, and the relative expression of p-I 魏 B protein and NF- 魏 B p65 protein increased with the increase of Flavatin concentration (P 0.05). Conclusion Acanthopanthin can inhibit the proliferation of human gastric cancer MKN-45 cell line and promote apoptosis. The mechanism may be the activation of NF- 魏 B signaling pathway by Acanthopanthin.
