氧化石墨烯介導的光熱免疫療法治療轉移性小鼠乳腺腫瘤

【摘要】:本研究分別從細胞層面和在體層面,研究納米氧化石墨烯(nano graphene oxide,NGO)聯(lián)合805 nm近紅外激光對于腫瘤的殺傷和免疫刺激作用.細胞實驗證實NGO具有良好的光熱轉換效應,聯(lián)合激光治療,能夠有效殺傷腫瘤細胞.此外,NGO能夠刺激巨噬細胞產生IL-6及TNFα,并增強激光殺傷的腫瘤細胞對巨噬細胞的免疫刺激效應.在體實驗中,在小鼠背部不同位置使用同一瘤株種植兩個瘤塊,模擬局部原發(fā)腫瘤和轉移瘤.實驗結果表明,NGO聯(lián)合激光治療可以有效消融局部腫瘤,并且,未治療的遠隔部位腫瘤生長速度也顯著降低.說明NGO聯(lián)合激光治療的方式在局部殺傷原位腫瘤的同時,可能誘發(fā)了機體的抗腫瘤免疫反應,從而達到了抑制遠隔腫瘤生長的效果.
[Abstract]:In this study, the cytotoxic and immunostimulatory effects of nano-graphene oxide (nano graphene oxide,NGO) combined with 805 nm near infrared laser on tumor were studied at the cellular and in vivo levels, respectively. Cell experiments have proved that NGO has a good photothermal conversion effect, combined with laser therapy, can effectively kill tumor cells. In addition, NGO stimulated macrophages to produce IL-6 and TNF 偽, and enhanced the immunostimulatory effect of laser killed tumor cells on macrophages. In vivo, two tumor blocks were implanted with the same tumor strain at different sites in the back of mice to simulate local primary tumor and metastatic tumor. The experimental results show that NGO combined with laser therapy can effectively ablation local tumors, and the growth rate of untreated distant tumors is also significantly reduced. The results suggest that the combination of NGO and laser therapy may induce antitumor immune response and inhibit the growth of distant tumor.
