【摘要】：目的： C12orf48是新近研究發(fā)現(xiàn)的一個(gè)基因,在其他惡性腫瘤中已有研究,但在胃癌中的表達(dá)尚未知曉,本文初步探討C12orf48基因在胃癌組織以及胃癌細(xì)胞系中的表達(dá),并將其表達(dá)和胃癌患者的臨床病理資料進(jìn)行相關(guān)性分析,探討其與患者臨床病理特征的相關(guān)性。方法： 采用Real time-PCR和Western-blot方法檢測(cè)C12orf48在胃癌細(xì)胞中的表達(dá)情況,應(yīng)用免疫組織化學(xué)檢查C12orf48在53例胃癌組織及對(duì)應(yīng)非癌胃組織中的表達(dá)。結(jié)果： 5株胃癌細(xì)胞中,C12orf48在未分化細(xì)胞系HGC的RNA表達(dá)水平顯著低于低分化胃癌細(xì)胞系A(chǔ)GS、KATO3、MGC和中等分化胃癌細(xì)胞系SGC7901,但C12orf48蛋白在HGC細(xì)胞中的表達(dá)水平則明顯增高,而在MGC細(xì)胞中蛋白水平則較低。在胃癌標(biāo)本中C12orf48陽性表達(dá)20例(37.7%),相應(yīng)非癌胃組織陽性41例(77.4%),差異有統(tǒng)計(jì)學(xué)意義(P=0.000)。 結(jié)論： C12orf48在胃癌細(xì)胞系中表達(dá)情況存在明顯差異,在胃癌組織中C12orf48的表達(dá)低于非癌胃組織,但和胃癌的浸潤(rùn)深度、淋巴結(jié)轉(zhuǎn)移、TNM分期以及腫瘤分化程度無相關(guān)性。
[Abstract]:Objective: C12orf48 is a newly discovered gene, which has been studied in other malignant tumors, but its expression in gastric cancer has not been known. In this paper, we studied the expression of C12orf48 gene in gastric cancer tissues and gastric cancer cell lines. The correlation between the expression and clinicopathological data of gastric cancer patients was analyzed to explore the correlation between the expression and clinicopathological features of gastric cancer patients. Methods: the expression of C12orf48 in gastric cancer cells was detected by Real time-PCR and Western-blot, and the expression of C12orf48 in 53 cases of gastric cancer and corresponding non-cancerous gastric tissues was detected by immunohistochemistry. Results: the RNA expression level of C12orf48 in undifferentiated gastric cancer cell line HGC was significantly lower than that in poorly differentiated gastric cancer cell line AGS,KATO3,MGC and moderately differentiated gastric cancer cell line SGC7901, but the expression level of C12orf48 protein in HGC cells was significantly increased. However, the protein level in MGC cells was lower. The positive expression of C12orf48 was found in 20 cases (37.7%) of gastric cancer and 41 cases (77.4%) of corresponding non-cancerous gastric tissues. The difference was statistically significant (P0. 000). Conclusion: there is significant difference in the expression of C12orf48 in gastric cancer cell line, and the expression of C12orf48 in gastric cancer is lower than that in non-cancerous gastric tissue, but there is no correlation between the expression of C12orf48 and the depth of invasion, lymph node metastasis and tumor differentiation.
