大鼠小膠質(zhì)細(xì)胞及巨噬細(xì)胞對(duì)膠質(zhì)瘤C6細(xì)胞株遷移能力的影響

【摘要】:目的探討大鼠小膠質(zhì)細(xì)胞及外周血巨噬細(xì)胞對(duì)膠質(zhì)瘤C6細(xì)胞株遷移能力的影響。方法將大鼠小膠質(zhì)細(xì)胞及外周血單核細(xì)胞誘導(dǎo)分化形成的巨噬細(xì)胞分別與膠質(zhì)瘤C6細(xì)胞共培養(yǎng),以大鼠成纖維細(xì)胞共培養(yǎng)為陽性對(duì)照,單純膠質(zhì)瘤C6細(xì)胞為空白對(duì)照,共培養(yǎng)0、24、48 h分別行劃痕實(shí)驗(yàn),于劃痕后6 h觀察劃痕實(shí)驗(yàn)結(jié)果;共培養(yǎng)24、48 h時(shí)以CD11b標(biāo)記、流式分選分離小膠質(zhì)/巨噬細(xì)胞及C6細(xì)胞,行Western blot檢測(cè)0、24、48 h小膠質(zhì)/巨噬細(xì)胞IL-10、IL-18和MMP-9及C6細(xì)胞株TGF-β、FAS配體表達(dá)水平的改變。結(jié)果早期小膠質(zhì)細(xì)胞及巨噬細(xì)胞均抑制膠質(zhì)瘤的遷移,巨噬細(xì)胞的抑制效果更強(qiáng)(P0.05);與膠質(zhì)瘤細(xì)胞共培養(yǎng)后兩種細(xì)胞對(duì)腫瘤的抑制作用消失且差異無統(tǒng)計(jì)學(xué)意義,均表現(xiàn)為促進(jìn)腫瘤遷移。小膠質(zhì)細(xì)胞、巨噬細(xì)胞在初始階段IL-10、IL-18、金屬基質(zhì)蛋白酶MMP-9表達(dá)略有不同,但共培養(yǎng)后兩種細(xì)胞的上述蛋白表達(dá)均升高且一致;共培養(yǎng)后的膠質(zhì)瘤細(xì)胞的TGF-β、FAS配體較共培養(yǎng)前表達(dá)均升高且一致。結(jié)論小膠質(zhì)細(xì)胞及巨噬細(xì)胞與C6膠質(zhì)瘤細(xì)胞共培養(yǎng)后被馴化成同樣的GAMs,對(duì)C6細(xì)胞遷移影響的生物學(xué)表現(xiàn)一致。
[Abstract]:Objective to investigate the effects of microglia and peripheral blood macrophages on the migration of glioma C6 cells. Methods Rat microglial cells and macrophages derived from peripheral blood monocytes were co-cultured with glioma C6 cells respectively. Rat fibroblasts were co-cultured as positive control and glioma C6 cells as blank control. The results of scratch test were observed at 6 h after scratch, and CD11b labeled with CD11b was used to separate microglia / macrophages and C6 cells at 48 h after co-culture, and microglia / macrophages and C6 cells were separated by flow cytometry. The expression of IL-18 and MMP-9 in microglia / macrophages and the expression of TGF- 尾 -FAS ligand in C6 cell line were detected by Western blot. Results both microglia and macrophages inhibited the migration of gliomas in the early stage, and the inhibitory effect of macrophages was stronger (P0.05), but the inhibitory effects of the two kinds of cells on glioma disappeared after co-culture with glioma cells, and there was no significant difference between the two kinds of cells. All of them were found to promote tumor migration. The expression of IL-10 IL-18 and MMP-9 was slightly different in microglia and macrophage at the initial stage, but the expression of these proteins was increased and consistent after co-culture. The expression of TGF- 尾 -FAS ligand in co-cultured glioma cells was higher and consistent than that before co-culture. Conclusion microglia and macrophages co-cultured with C6 glioma cells were acclimated into the same GAMs.The biological effects on the migration of C6 cells were consistent.
