【摘要】:目的:本研究通過探索miR-125b-1-3p在胃癌及對應(yīng)癌旁組織中的表達(dá)情況,并分析其與胃癌臨床病理特征之間的關(guān)系,從而探討miR-125b-1-3p作為胃癌診斷標(biāo)志物和治療靶點(diǎn)的可能性。方法:實(shí)驗(yàn)前期利用基因芯片技術(shù)檢測14例早癌患者的胃癌組織及對應(yīng)癌旁組織中miRNAs的表達(dá)譜分析情況。收集42例手術(shù)患者的胃癌組織及對應(yīng)的癌旁組織。首先在胃癌樣本及癌旁樣本中提取總的RNA,運(yùn)用實(shí)時(shí)熒光定量PCR檢測出miR-125b-1-3p在胃癌組織中表達(dá)水平,并分析miR-125b-1-3p的表達(dá)水平與胃癌臨床病理特征之間的關(guān)系。結(jié)果:實(shí)時(shí)熒光定量PCR結(jié)果提示胃癌組織miR-125b-1-3p的表達(dá)相較于對應(yīng)癌旁組織存在顯著下調(diào),有統(tǒng)計(jì)學(xué)差異。miR-125b-1-3p在胃癌的低表達(dá)僅與分化程度存在相關(guān)性,而與性別、TNM分期、浸潤深度、淋巴結(jié)轉(zhuǎn)移、年齡、腫瘤大小等胃癌臨床病理特征均無明顯相關(guān)性。在miR-125b-1-3p低表達(dá)的胃癌組織中,根據(jù)ROC曲線確定胃癌組織高/中分化程度的曲線下面積是0.714(P0.05),得出高/中分化程度的最佳界值:相對表達(dá)量為0.18,其在診斷高/中分化程度的敏感度70%、特異性72.2%、陽性預(yù)測值70%、陰性預(yù)測值30%。當(dāng)相對表達(dá)量≥0.18時(shí)發(fā)生高/中分化程度的風(fēng)險(xiǎn)是相對表達(dá)量0.18時(shí)患者的2.33倍(P0.05),提示相對表達(dá)量≥0.18是胃癌組織高/中分化程度的獨(dú)立預(yù)測因素。結(jié)論:miR-125b-1-3p在胃癌組織中顯著下調(diào),當(dāng)miR-125b-1-3p的相對表達(dá)量≥0.18在預(yù)測胃癌高/中分化程度上有較高的診斷價(jià)值。
[Abstract]:Objective: to explore the expression of miR-125b-1-3p in gastric cancer and its relationship with the clinicopathological features of gastric cancer, and to explore the possibility of miR-125b-1-3p as a diagnostic marker and a therapeutic target for gastric cancer. Methods: the expression profiles of miRNAs in gastric cancer tissues and adjacent tissues of 14 patients with early cancer were detected by microarray technique. Gastric cancer tissues and corresponding paracancerous tissues were collected from 42 patients. First, the total RNAs were extracted from gastric cancer samples and adjacent samples. The expression of miR-125b-1-3p in gastric cancer tissues was detected by real-time fluorescent quantitative PCR, and the relationship between the expression level of miR-125b-1-3p and clinicopathological features of gastric cancer was analyzed. Results: the results of real-time fluorescence quantitative PCR showed that the expression of miR-125b-1-3p in gastric cancer was significantly down-regulated compared with the corresponding adjacent tissues, and the low expression of miR-125b-1-3p in gastric cancer was only correlated with the degree of differentiation, but with the TNM stage and depth of invasion. The clinicopathological features of gastric cancer, such as lymph node metastasis, age and tumor size, were not significantly correlated. In gastric cancer tissues with low expression of miR-125b-1-3p, According to the ROC curve, the area under the curve of high / medium differentiation degree of gastric cancer is 0.714 (P0.05). The best boundary value of high / medium differentiation degree is obtained: the relative expression is 0.18, its sensitivity in diagnosing high / medium differentiation degree is 70, specificity 72.2, positive.
