文章摘要:腫瘤干細(xì)胞學(xué)說(shuō)認(rèn)為大部分腫瘤來(lái)源于腫瘤干細(xì)胞。腫瘤干細(xì)胞與正常干細(xì)胞一樣具有自我更新能力。能夠產(chǎn)生腫瘤組織團(tuán)塊中的各種增殖和分化的細(xì)胞。腫瘤干細(xì)胞可能來(lái)源于正常干細(xì)胞或分化細(xì)胞。干細(xì)胞niche是干細(xì)胞生存的微環(huán)境.通過(guò)提供抑制細(xì)胞增殖和生長(zhǎng)的信號(hào)來(lái)維持干細(xì)胞于靜止?fàn)顟B(tài)。干細(xì)胞niche功能異常會(huì)導(dǎo)致niche的數(shù)量增加.以及干細(xì)胞功能異常和數(shù)量增加。腫瘤干細(xì)胞可能在異常的niche中存活,打破這種異常的niche就會(huì)削弱腫瘤干細(xì)胞的自我更新.從而抑制腫瘤的生長(zhǎng)。靶向腫瘤干細(xì)胞異常微環(huán)境的治療策略可能是癌癥治療的一個(gè)方向。
Abstr:The tumor stem cell model proposes that tumor stem cells are the origin of most tumors. Like normal stem cells, tumor stem cells have the ability to self-renew and to give rise to the variety of proliferating and differentiated cells that make up the bulk of a tumor, Tumor stem cells could be derived from normal stem cells or differentiated cells. The stem cell niches are the microenvironment that maintains stem ceils primarily in a quiescent state by providing signals that inhibit cell proliferation and growth. Loss of niches can result in loss of stem cells and increasing niches lead to an increased number of stem cells. Tumor stem cells could live in abnormal niches. Disrupting this abnormal niches may impair tumor stem cell self- renewal, thereby, significantly inhibits the growth of tumors. Targeting the unique microenvironment of tumor stem cells may be the key of effective cancer therapy.
