【摘要】：目的:研究Runx2對肝癌細胞中MMP9的表達以及其對肝癌細胞侵襲遷移能力的影響。方法:通過免疫組織化學染色法對2005年12月至2015年12月天津醫(yī)科大學腫瘤醫(yī)院和天津醫(yī)科大學總醫(yī)院189例肝細胞肝癌病例標本中Runx2和MMP9的表達進行分析;將Runx2過表達質(zhì)粒和干擾質(zhì)粒分別轉(zhuǎn)染至肝癌細胞系HepG2和SMMC7721細胞中。使用Western blot法檢測轉(zhuǎn)染后HepG2和SMMC7721細胞中Runx2和MMP9的表達情況;劃痕、侵襲實驗檢測Runx2對肝癌細胞遷移和侵襲能力的影響。結(jié)果:免疫組織化學結(jié)果顯示Runx2的表達和MMP9的表達相關(guān)。在HepG2細胞中上調(diào)Runx2的表達,促進了MMP9的表達,增強了HepG2細胞遷移侵襲能力;在SMMC7721細胞中下調(diào)Runx2的表達,降低了MMP9的表達,抑制了SMMC7721細胞的遷移侵襲能力。結(jié)論:Runx2的表達可能參與促進肝細胞肝癌細胞MMP9的表達,進而促進肝細胞肝癌的遷移侵襲能力。
[Abstract]:Aim: to study the effect of Runx2 on the expression of MMP9 and the ability of invasion and migration of hepatoma cells. Methods: from December 2005 to December 2015, the expression of Runx2 and MMP9 in 189 cases of hepatocellular carcinoma (HCC) in Tianjin Medical University Oncology Hospital and Tianjin Medical University General Hospital were analyzed by immunohistochemical staining. Runx2 overexpression plasmid and interference plasmid were transfected into hepatoma cell lines HepG2 and SMMC7721, respectively. The expression of Runx2 and MMP9 in transfected HepG2 and SMMC7721 cells was detected by Western blot assay, and the effect of Runx2 on the migration and invasion of hepatoma cells was detected by scratch and invasion assay. Results: immunohistochemical results showed that the expression of Runx 2 was correlated with the expression of MMP9. The expression of Runx2 was up-regulated in HepG2 cells, which promoted the expression of MMP9 and enhanced the ability of migration and invasion of HepG2 cells, and down-regulated the expression of Runx2 in SMMC7721 cells, reduced the expression of MMP9 and inhibited the migration and invasion of SMMC7721 cells. Conclusion the expression of 1: Runx2 may play an important role in promoting the expression of MMP9 in hepatocellular carcinoma cells, thus promoting the ability of migration and invasion of hepatocellular carcinoma.
