腦源性神經(jīng)營養(yǎng)因子-反義長鏈非編碼RNA過表達可抑制肝癌細胞的增殖和侵襲

【摘要】:目的:探討腦源性神經(jīng)營養(yǎng)因子-反義長鏈非編碼RNA(brain-derived neurotrophic factor-antisense long non-coding RNA,BDNF-AS)對人肝癌細胞增殖和侵襲能力的影響。方法:將攜帶BDNF-AS基因的重組慢病毒分別感染肝癌細胞系HepG2和97-H后,采用RT-PCR法檢測肝癌細胞中BDNF-AS和BDNF mRNA的表達水平變化。分別采用EdU摻入實驗、AnnexinⅤ-FITC/碘化丙啶(propidium iodide,PI)雙染和Transwell小室法檢測BDNF-AS過表達對HepG2和97-H細胞增殖、凋亡和侵襲能力的影響。結(jié)果:感染攜帶BDNF-AS基因的重組慢病毒后,肝癌HepG2和97-H細胞中BDNF-AS的表達水平均明顯升高(P值均0.000 1),而BDNF mRNA的表達水平明顯降低(P0.000 1,P=0.001)。BDNF-AS過表達后,HepG2和97-H細胞的凋亡率無明顯變化(P值均0.05),但細胞增殖率明顯降低(P0.05,P=0.001),侵襲細胞數(shù)明顯減少(P值均0.000 1)。結(jié)論:BDNF-AS過表達可能對肝癌細胞的生長和侵襲具有重要的負調(diào)控作用。
[Abstract]:Aim: to investigate the effects of brain-derived neurotrophic factor-antisense long chain noncoding RNA(brain-derived neurotrophic factor-antisense long non-coding RNAs BDNF-ASon on proliferation and invasion of human hepatoma cells. Methods: after the recombinant lentivirus carrying BDNF-AS gene was infected with HepG2 and 97-H, the expression levels of BDNF-AS and BDNF mRNA in HCC cells were detected by RT-PCR assay.
