FOXC2對肺腺癌細胞惡性增殖及侵襲遷移能力的影響

【摘要】:目的探究叉頭框C2蛋白(FOXC2)對肺腺癌細胞惡性增殖及侵襲遷移能力的影響。方法利用慢病毒轉(zhuǎn)染的方法在人肺腺癌細胞系A549中構(gòu)建過表達FOXC2細胞(實驗組)及相應對照細胞系(對照組),Western印跡檢測兩組細胞FOXC2及干性相關分子Oct4、Sox2及Nanog的表達水平,MTT法檢測兩組細胞0、24、48、72、96 h的惡性增殖能力,Transwell侵襲及遷移實驗檢測兩組細胞侵襲遷移能力。結(jié)果與對照組細胞比較,實驗組細胞FOXC2、Oct4、Sox2及Nanog表達水平均顯著上調(diào)(P0.05),細胞惡性增殖能力在48、72及96 h出現(xiàn)顯著升高(P0.05),細胞侵襲及遷移能力均顯著上調(diào)(P0.05),差異均有統(tǒng)計學意義。結(jié)論 FOXC2可促進肺腺癌細胞惡性增殖及侵襲遷移能力,其機制與介導腫瘤細胞干細胞特性形成有關。
[Abstract]:Objective to investigate the effects of forked C2 protein (FOXC2) on the proliferation and invasion and migration of lung adenocarcinoma cells. Methods the expression of FOXC2 cells (experimental group) and corresponding control cell lines (control group) in human lung adenocarcinoma cell line A549 were constructed by lentivirus transfection. The Western blot was used to detect FOXC2 and Oct4 in two groups of cells. The expression level of Sox2 and Nanog, MTT method was used to detect the malignant proliferation ability of 0,24,48,72,96 h in two groups of cells. Transwell invasion and migration test was used to detect the invasion and migration of cells in the two groups. Results compared with the control group, the expression level of FOXC2, Oct4, Sox2 and Nanog in the experimental group were up (P0.05), and the cell malignant proliferation ability was in 48,72 and 96 Significant increase (P0.05), cell invasion and migration ability were significantly up (P0.05), and the difference was statistically significant. Conclusion FOXC2 can promote the malignant proliferation and invasion and migration of lung adenocarcinoma cells, and the mechanism is related to the formation of tumor cell stem cell characteristics.
