基于超聲波穿孔效應的細胞SERS光譜研究

【摘要】:本論文闡述了超聲波穿孔效應的細胞SERS光譜研究。針對現(xiàn)有將金屬納米粒子導入活細胞中方法上存在的缺陷,我們首次開展了超聲波穿孔效應-SERS光譜技術研究,即通過超聲波穿孔效應將金屬納米顆粒(金和銀)快速(1分鐘)導入活細胞中作為SERS增強基底,獲得細胞的SERS光譜,從而實現(xiàn)對細胞內(nèi)各物質(zhì)生化成分的研究及多種癌細胞的檢測和篩查應用。主要研究成果如下:1、利用超聲波穿孔效應將銀納米顆?焖賹隟562、NP69及C666細胞,結合超聲波穿孔后的細胞TEM圖像驗證了超聲波穿孔效應確實能夠使得金屬納米顆?焖龠M入細胞內(nèi);2、通過細胞超聲波穿孔效應-SERS光譜的分析獲得了最佳的優(yōu)化測試條件:采用20倍物鏡并將光斑擴束到100%;3、利用超聲波穿孔效應法與傳統(tǒng)孵育內(nèi)吞法下獲得全細胞SERS光譜存在很大差異,可能跟銀納米顆粒具有毒性作用有關;4、通過超聲波穿孔效應-SERS技術獲得的正常鼻咽NP69細胞與鼻咽癌C666細胞的SERS光譜,從而進行癌癥細胞的篩查研究;其次,用同樣方法對比分析NP69與K562細胞;最后再進行三種細胞的區(qū)分篩查與分析。實驗結果表明,超聲波穿孔效應-SERS技術方法可以在1分鐘內(nèi)將金和銀納米顆?焖賹牖罴毎麅(nèi)部并作為SERS增強基底,通過檢測細胞SERS光譜,可快速確定細胞的生化成分,并應用于癌細胞的篩查研究。該方法不僅具有操作簡便快速、分辨率高等優(yōu)點,而且擴展了SERS技術在細胞生化研究領域的應用,其應用前景十分廣闊。
[Abstract]:In this paper, the cell SERS spectroscopy of ultrasonic perforation is described. In view of the defects of the existing methods for the introduction of metal nanoparticles into living cells, we have carried out the first research on ultrasonic perforation effect (SERS) spectroscopy. In other words, the metal nanoparticles (gold and silver) were introduced into living cells as SERS enhancement substrate by ultrasonic piercing effect for 1 minute, and the SERS spectra of the cells were obtained. Thus, the biochemical components of various substances in the cell and the detection and screening of many kinds of cancer cells can be realized. The main results are as follows: 1. Silver nanoparticles were rapidly introduced into K562NP-69 and C666 cells by ultrasonic perforation. Combined with the TEM images of cells after ultrasonic perforation, it was proved that the ultrasonic perforation effect could make metal nanoparticles enter the cell rapidly. The best optimized measurement was obtained by the analysis of cell ultrasonic perforation effect -SERS spectra. The test conditions were as follows: using 20 times objective lens and expanding the light spot to 100 ~ 3, the SERS spectra of whole cell obtained by ultrasonic perforation method and traditional incubation endocytosis method were very different. It may be related to the toxic effect of silver nanoparticles. The SERS spectra of normal nasopharyngeal NP69 cells and nasopharyngeal carcinoma C666 cells obtained by ultrasonic perforation effect -SERS technique were used to screen cancer cells. NP69 and K562 cells were compared with each other by the same method. The experimental results show that the ultrasonic perforation effect -SERS technique can quickly transfer gold and silver nanoparticles into living cells within one minute and act as a SERS enhancement substrate. The biochemical components of the cells can be quickly determined by detecting the SERS spectra of the cells. And applied to the screening of cancer cells. This method not only has the advantages of simple and fast operation and high resolution, but also extends the application of SERS technology in the field of cell biochemistry, and its application prospect is very broad.
