【摘要】：目的探討miR-551b在人胃癌組織中的表達及對胃癌細胞凋亡的影響。方法采用Real-time PCR檢測胃癌組織、正常胃組織miR-551b表達水平,分別將miRNA無義序列、miR-551bmimics、miR-551binhibitors轉(zhuǎn)染至人胃癌細胞株SGC-7901,Real-time PCR檢測各組細胞miR-551b表達,MTT法檢測各組腫瘤細胞增殖活性,Transwell法檢測各組細胞侵襲能力,流式細胞術檢測各組細胞凋亡情況,Hoechst33342熒光染色觀察各組細胞自噬、凋亡的發(fā)生,Western blot檢測各組細胞NF-κB、LC3Ⅱ、Beclin 1蛋白表達。結(jié)果 miR-551b在胃癌組織表達明顯下調(diào),胃癌組織miR-551b相對表達量(1.75±0.13)顯著低于正常胃組織(2.47±0.38)(P0.05)。與NC組和miR-551binhibitors組比較,miR-551bmimics組miR-551b表達水平顯著上升,細胞增殖率和侵襲率顯著降低,細胞凋亡率明顯上升,各指標比較差異均具有統(tǒng)計學意義(均P0.05)。Hoechst33342熒光染色顯示miR-551bmimics組出現(xiàn)大量自噬泡,NC組可見部分自噬泡,而miR-551binhibitors組僅有少量自噬泡。Western blot檢測結(jié)果顯示,與NC組和miR-551binhibitors組比較,miR-551bmimics組NF-κB、LC3Ⅱ、Beclin 1蛋白表達水平顯著升高(均P0.05)。結(jié)論 miR-551b能夠抑制胃癌細胞的增殖和侵襲等細胞生物功能,其作用機制可能是誘導了胃癌細胞發(fā)生自噬性凋亡。
[Abstract]:Objective to investigate the expression of miR-551b in human gastric carcinoma and its effect on the apoptosis of gastric cancer cells. Methods Real-time PCR was used to detect the expression of miR-551b in gastric cancer and normal gastric tissues. MiRNA nonsense sequence was transfected into human gastric cancer cell line SGC-7901Real-time PCR. MiR-551b expression in each group was detected. The proliferation activity of tumor cells in each group was detected by miRNA method and the invasiveness of tumor cells in each group was detected by Transwell method.
