馬來酸噻嗎洛爾對血管瘤動物模型雞冠血管損傷效應

【摘要】:目的研究毛細血管瘤模型雞冠經(jīng)馬來酸噻嗎洛爾滴眼液處理后組織病理學變化以及血管內(nèi)皮細胞凋亡情況,初步探討馬來酸噻嗎洛爾局部濕敷對毛細血管的損傷效應及聯(lián)合聚桂醇注射治療提高聚桂醇治療效果的可行性。方法取6月齡健康雄性雞30只,隨機分為五組,每組6只.對照組E(0.9%的氯化鈉溶液外用)。實驗組:A組用馬來酸噻嗎洛爾滴眼液外用;B組用醋酸曲安奈德注射液外用;C組予以聚桂醇注射液注射治療;D組馬來酸噻嗎洛爾滴眼液與聚桂醇注射液聯(lián)合應用。觀察各組經(jīng)藥物處理后動物模型的一般情況,分別于處理后第7天,14天,21天,28天從ABCDE五個組隨機選取一只雞,取材后,10%福爾馬林溶液固定,常規(guī)HE染色,在光學顯微鏡下選5個高倍鏡視野(10X40倍)并拍攝照片,統(tǒng)計毛細血管數(shù)目,計算其平均值,并行TUNEL染色觀察毛細血管內(nèi)皮細胞凋亡情況。結(jié)果實驗組雞冠與對照組相比,其顏色均有不同程度變淺變淡;HE染色切片光鏡下觀察可見毛細血管數(shù)目減少,與E組相比,差異均有統(tǒng)計學意義(p均0.05);行TUNEL染色觀察可見噻嗎洛爾干預組真皮乳頭層凋亡毛細血管內(nèi)皮細胞,聚桂醇干預組真皮網(wǎng)狀層可見少量凋亡毛細血管內(nèi)皮細胞,并且隨著觀察周期的延長,凋亡細胞數(shù)目逐漸增多,E組與A,B,C,D組相比較,毛細血管凋亡數(shù)目差異均有統(tǒng)計學意義(p均0.05)。結(jié)論馬來酸噻嗎洛爾可誘導血管內(nèi)皮細胞凋亡,毛細血管數(shù)目減少。馬來酸噻嗎洛爾與聚桂醇聯(lián)合應用可起協(xié)同作用,可致管腔縮窄,血管壁結(jié)構(gòu)消失,損傷血管內(nèi)皮并誘導毛細血管內(nèi)皮細胞發(fā)生凋亡,并且不會造成雞冠外形的改變,符合無創(chuàng)治療血管瘤要求,為血管瘤的非手術(shù)治療提供理論依據(jù)和治療參考數(shù)據(jù)。
[Abstract]:Objective to study the histopathological changes and the apoptosis of vascular endothelial cells (VEC) in the model of capillary hemangioma treated with tiamol maleate eye drops. To explore the effect of local wet compress of tiamolol maleate on capillary injury and the feasibility of combined injection of polycinnamol to improve the efficacy of polycinnamol. Methods 30 healthy male chickens aged 6 months were randomly divided into five groups. There were 6 rats in each group. The control group was given 0.9% sodium chloride solution for external use. Experimental group A was treated with tiamolol maleate eyedrops. Group B was treated with triamcinolone acetonide acetate injection and group C was injected with polycinnamol maleate. To observe the general situation of the animal model after drug treatment. A chicken was randomly selected from five ABCDE groups on the 7th day, 14th day, 21st day and 28th day after treatment. The chicken was fixed in 10% formalin solution, stained with HE, and 5 high power field of vision was selected under optical microscope (10 x 40 times). The number of capillaries was counted, the average of capillaries was calculated, and the apoptosis of capillary endothelial cells was observed by TUNEL staining. The number of capillaries was decreased under light microscope, compared with E group. The results showed that the apoptosis of capillary endothelial cells in dermal papilla was observed by TUNEL staining, and a small number of apoptotic capillary endothelial cells were observed in dermal reticular layer of polycinnamol treated group. Moreover, the number of apoptotic cells increased gradually with the prolongation of the observation cycle. There were significant differences in the number of apoptotic capillaries between group E and group A (P < 0.05). Conclusion tiamolol maleate can induce apoptosis of vascular endothelial cells. The combination of thiamolol maleate and polycinnamol may play a synergistic role in constricting the lumen, disappearing the vascular wall structure, damaging the vascular endothelium and inducing the apoptosis of the capillary endothelial cells. And it does not cause the change of the shape of chicken crown, which accords with the requirement of noninvasive treatment of hemangioma, and provides the theoretical basis and treatment reference data for the non-surgical treatment of hemangioma.
