細(xì)胞周期相關(guān)蛋白在人結(jié)腸癌組織中的表達(dá)及其臨床意義的探討

【摘要】:目的:檢測結(jié)腸癌患者組織中細(xì)胞周期蛋白D1(cyclin D1)、細(xì)胞周期蛋白E(cyclin E)以及NIK-IKK結(jié)合蛋白(NIBP)的表達(dá),探討其與臨床、病理特征之間的關(guān)系,并分析三者在結(jié)腸癌組織中表達(dá)的相關(guān)性。為結(jié)腸癌的早期診斷和預(yù)后提供一個可靠的分子生物學(xué)指標(biāo)。方法:收集從2015年2月到2015年12月之間廣西醫(yī)科大學(xué)第一附屬醫(yī)院結(jié)直腸外科經(jīng)外科手術(shù)切除及消化內(nèi)科腸鏡活檢的部分標(biāo)本130例,正常結(jié)腸黏膜、結(jié)腸癌各65例,所有的患者均未接受放化療。采用免疫組織化學(xué)SP法分別檢測并分析NIBP、cyclin E和cyclinD1的表達(dá)和相關(guān)性,并分析上述蛋白的表達(dá)與結(jié)腸癌患者腫瘤的浸潤深度、TNM臨床分期、淋巴結(jié)轉(zhuǎn)移和遠(yuǎn)處轉(zhuǎn)移相關(guān)性。結(jié)果:(1)NIBP在結(jié)腸癌組織中高表達(dá)陽性率為60.00%(39/65),明顯高于正常結(jié)腸粘膜組織中的陽性表達(dá)率[18.46%(12/65),P0.05],差異具有統(tǒng)計學(xué)意義;Cyclin E在結(jié)腸癌組織中高表達(dá)陽性率為56.92%(37/65),顯著高于正常結(jié)腸粘膜組織中的陽性表達(dá)率[3.08%(2/65),P0.05],差異具有統(tǒng)計學(xué)意義;CyclinD1在結(jié)腸癌組織中高表達(dá)陽性率為89.23%(58/65),顯著高于正常結(jié)腸粘膜組織中的陽性表達(dá)率[43.08%(28/65),P0.05],差異具有統(tǒng)計學(xué)意義。(2)結(jié)腸癌組織中CyclinE、NIBP和Cyclin D1的表達(dá)與腫瘤浸潤深度、TNM臨床分期、淋巴結(jié)轉(zhuǎn)移和遠(yuǎn)處轉(zhuǎn)移明顯相關(guān)(p0.05)。(3)結(jié)腸癌組織中NIBP和Cyclin D1表達(dá)之間呈正相(r=0.32,P0.05),NIBP和CyclinE表達(dá)之間呈正相關(guān)(r=0.43,P0.05),CyclinD1和CyclinE表達(dá)之間呈正相關(guān)(r=0.23,P0.05)。結(jié)論:(1)NIBP、Cyclin E、Cyclin D1在結(jié)腸癌中高表達(dá),三者可能參與了結(jié)腸癌的發(fā)生發(fā)展;(2)三者在結(jié)腸癌中的高表達(dá)都可促進(jìn)腫瘤細(xì)胞的浸潤與轉(zhuǎn)移,其表達(dá)越高,浸潤深度越深,淋巴結(jié)及遠(yuǎn)處轉(zhuǎn)移可能性越大。(3)NIBP可能促進(jìn)CyclinE和Cyclin D1的高表達(dá)而導(dǎo)致細(xì)胞周期調(diào)控異常,三者可能共同參與了結(jié)腸癌的發(fā)生發(fā)展。
[Abstract]:Objective: to detect cyclin D1 cyclin D1 in the tissues of colon cancer patients. The expression of cyclin E (cyclin E) and NIK-IKK binding protein (NIBP) and its relationship with clinical and pathological features were investigated. The correlation of the expression of the three proteins in colon cancer tissues was analyzed to provide a reliable molecular biological index for the early diagnosis and prognosis of colon cancer. Methods:. A total of 130 specimens were collected from the first affiliated Hospital of Guangxi Medical University from February 2015 to December 2015. 65 cases of normal colon mucosa and 65 cases of colon cancer, all patients did not receive radiotherapy and chemotherapy. Immunohistochemical SP method was used to detect and analyze NIBP. The expression and correlation of cyclin E and cyclinD1, and the relationship between the expression of these proteins and the depth of tumor invasion in patients with colon cancer were analyzed. Results the positive rate of high expression of NIBP in colon cancer tissues was 60.009 / 65). The positive expression rate in normal colonic mucosa was significantly higher than that in normal colonic mucosa. [18.46% / 65% P 0.05, the difference was statistically significant; The positive rate of high expression of Cyclin E in colon cancer tissues was 56.92% 37 / 65, which was significantly higher than that in normal colonic mucosa tissues. [The difference was statistically significant (P 0.05). The high expression rate of CyclinD1 in colon cancer tissue was 89.23%, which was significantly higher than that in normal colonic mucosa tissue. [43.08% 28% 65% (P0.05), the difference was statistically significant (P < 0.05) CyclinE in colon cancer tissue. The expression of NIBP and Cyclin D1 was correlated with the depth of tumor invasion. Lymph node metastasis and distant metastasis were significantly correlated with the expression of NIBP and Cyclin D1 in colorectal cancer tissues. There was a positive correlation between the expression of NIBP and CyclinE and the expression of cyclin D1 and CyclinE. Conclusion the high expression of Cyclin E Cyclin D1 in colon cancer may be involved in the occurrence and development of colon cancer. (2) the high expression of these three proteins in colon cancer can promote the invasion and metastasis of tumor cells, the higher the expression, the deeper the depth of invasion. The higher the possibility of lymph node and distant metastasis, the higher the expression of CyclinE and Cyclin D1, which may lead to abnormal cell cycle regulation. The three may be involved in the occurrence and development of colon cancer.
