模擬失重降低大鼠心肌細胞對異丙腎上腺素的反應性

【摘要】:本文旨在研究模擬失重對大鼠單個心肌細胞無負荷收縮功能的影響以及對異丙腎上腺素(isoproterenol,ISO)反應性的變化。采用大鼠尾部懸吊法在地面模擬失重狀態(tài),4周后以膠原酶Ⅰ消化分離心肌細胞,分別對左、右兩心室心肌細胞進行收縮功能測量。結(jié)果顯示,懸吊4周大鼠(懸吊組)左、右心室心肌細胞的長度和寬度與正常大鼠(對照組)相比均無顯著差異。隨刺激頻率增加,對照組與懸吊組大鼠心肌細胞縮短幅值均逐步增加。在1.0、2.0與4.0 Hz刺激下,對照組大鼠左心室心肌細胞縮短幅值分別為(8.50±1.26)%、(9.00±1.38)%與(9.23±1.83)%,右心室心肌細胞縮短幅值分別為(9.80±2.48)%、(10.03±2.48)%與(10.28±2.27)%:與對照組大鼠相比,在1.0與2.0 Hz刺激下,懸吊組大鼠左心室心肌細胞無負荷縮短幅值分別降低12.2%、10.9%(P0.05),右心室則分別降低16.5%、16.3%(P0.05);但是在4.0 Hz刺激下卻無顯著性改變。與同一頻率刺激下的對照組大鼠相比,懸吊組大鼠左、右心室心肌細胞達到縮短峰值的時程(time to peak shortening,TPS)明顯縮短(P0.05);而從縮短峰值至75%舒張的時程(TR_(75))則明顯延長(P0.05)。在各刺激頻率下,懸吊組大鼠左、右心室心肌細胞縮短(+dL/dt_(max))與舒張(-dL/dt_(max))速度均未發(fā)生明顯改變。用1、5、10 nmol/L ISO灌流達穩(wěn)態(tài)水平后,對照組大鼠心肌細胞縮短幅值分別增加了(10.63±0.83)%、(35.06±5.22)%和(71.64±6.83)%;而懸吊組大鼠心肌細胞縮短幅值僅增加(5.75±0.76)%、(23.97±4.50)%和(26.38±8.13)%,均有顯著性差異(P0.05,P0.01)。用10、50、100 nmol/L forskolin灌流達穩(wěn)定水平后,對照組大鼠心肌細胞縮短幅值分別增加了(3.04±0.27)%、(9.81±2.66)%、(20.20±3.47)%;而懸吊組大鼠心肌細胞縮短幅值僅增加了(1.42±0.53)%、(3.83±1.71)%、(5.49±4.08)%,均有顯著性差異(P0.05)。以上結(jié)果表明,模擬失重4周降低大鼠心肌細胞無負荷縮短幅值以及對ISO的反應性。
